In ovarian cancer, MEOX1 activates the SPHK1/S1P pathway to promote both tumor progression and tumor–stroma crosstalk. MEOX1‐dependent signaling drives CAF activation, enhances VEGF‐C expression, and stimulates lymphangiogenesis, ultimately facilitating lymph node metastasis.

Glioblastoma (GBM) exhibits RFX7 silencing via promoter hypermethylation, which downregulates PIK3IP1 to activate PI3K/AKT signaling, increase lactate accumulation, and enhance histone H4K12 lactylation. ARHGEF3 is broadly downregulated across human cancers and correlates with patient prognosis. Tumor‐intrinsic ARHGEF3 activates the RHOA–ROCK–PTEN cascade to inhibit AKT signaling, thereby promoting chemokine‐driven T‐cell infiltration and relieving lipid‐mediated myeloid immunosuppression.
