Mechanistic modelling of a recombinase-based two-input temporal logic gate [PDF]

open access: yes, 2017
Site-specific recombinases (SSRs) mediate efficient manipulation of DNA sequences in vitro and in vivo. In particular, serine integrases have been identified as highly effective tools for facilitating DNA inversion, enabling the design of genetic ...

Delivery of Islatravir via High Drug‐Load, Long‐acting Microarray Patches for the Prevention or Treatment of Human Immunodeficiency Virus

open access: yesAdvanced Healthcare Materials, Volume 14, Issue 7, March 14, 2025.
Islatravir‐loaded dissolving microarray patches (MAPs) provide a minimally invasive, long‐acting solution for HIV‐1 prevention and treatment. In rats, sustained islatravir delivery lasts up to three months, while in minipigs, efficacious plasma levels are detected for six days.

A Family of Developmentally Excised DNA Elements in \u3cem\u3eTetrahymena\u3c/em\u3e is under Selective Pressure to Maintain an Open Reading Frame Encoding an Integrase-Like Protein [PDF]

open access: yes, 2000
Tlr1 is a member of a family of ~20-30 DNA elements that undergo developmentally regulated excision during formation of the macronucleus in the ciliated protozoan Tetrahymena.

Unraveling the genomic mosaic of a ubiquitous genus of marine cyanobacteria [PDF]

open access: yes, 2008
Background: The picocyanobacterial genus Synechococcus occurs over wide oceanic expanses, having colonized most available niches in the photic zone. Large scale distribution patterns of the different Synechococcus clades (based on 16S rRNA gene markers)

Direct Access to α,β‐Unsaturated γ‐Lactams via Palladium‐Catalysed Carbonylation of Propargylic Ureas.

open access: yesAdvanced Synthesis &Catalysis, Volume 367, Issue 5, March 4, 2025.
Abstract Additive carbonylations typically necessitate strong nucleophiles, such as alcohols or amines. In this study, we carbonylated a poorly nucleophilic urea, under oxidant‐free conditions. Our straightforward carbonylative strategy enables access to versatile α,β‐unsaturated γ‐lactams featuring an aminocarbonyl fragment, utilizing readily ...

HIV-1 Integrase Strand Transfer Inhibitors with Reduced Susceptibility to Drug Resistant Mutant Integrases

open access: yesACS Chemical Biology, 2016
HIV integrase (IN) strand transfer inhibitors (INSTIs) are among the newest anti-AIDS drugs; however, mutant forms of IN can confer resistance. We developed noncytotoxic naphthyridine-containing INSTIs that retain low nanomolar IC50 values against HIV-1 ...

A simplified mathematical model of directional DNA site-specific recombination by serine integrases

open access: yesJournal of the Royal Society Interface, 2017
Serine integrases catalyse site-specific recombination to integrate and excise bacteriophage genomes into and out of their host's genome. These enzymes exhibit remarkable directionality; in the presence of the integrase alone, recombination between attP ...

Multiple choices for HIV therapy with integrase strand transfer inhibitors

open access: yesRetrovirology, 2012
Two integrases inhibitors, raltegravir and elvitegravir, have now been approved by regulatory agencies for use in the treatment of HIV-infected patients; and the approval of a third such drug, dolutegravir, is expected during 2013 on the basis of several

IS21 family transposase cleaved donor complex traps two right-handed superhelical crossings

open access: yesNature Communications, 2023
Transposases are ubiquitous enzymes that catalyze DNA rearrangement events with broad impacts on gene expression, genome evolution, and the spread of drug-resistance in bacteria.

Insights into the Dynamics and Binding Mechanisms of the Alkhumra Virus NS2B/NS3 Protease: A Molecular Dynamics Study

open access: yesAdvanced Theory and Simulations, EarlyView.
The ΔRMSF$\Delta {\rm RMSF}$ analysis reveals significant flexibility differences between free NS3 and the NS2B/NS3 complex, with notable deviations in specific regions. Key residues driving NS2B binding are identified, and the protonation state of catalytic serine affects oxyanion hole formation.
