The Pyrrolo[2,1-a]isoquinoline Alkaloids

The present chapter describes isolation, biogenetic proposals, and syntheses of the natural products 1-4 and 10-11 with a pyrrolo[2,1-a]-isoquinoline framework. Moreover, the syntheses of some structural analogs are discussed. The pyrrolo[2,1-a]isoquinolines are of interest due to their promising biological activities.

Abstract The use of chiral formamidines affixed to variously substituted tetrahydroisoquinolines, allows asymmetric C-C bond forming reactions to occur α- to the amino group. In this manner, a wide variety of (S)-1-alkyl-1,2,3,4-tetrahydroisoquinolines were constructed in > 90% enantiomeric excess.

Two new protoberberine alkaloids, 2,3,9,10-dimethylenedioxy-8-oxoprotoberberine (1) and 2,3,9,10-dimethylenedioxy-1,8-dihydroxyprotoberberine (2), together with nine known isoquinoline-type alkaloids were isolated from the roots of Thalictrum delavayi. Their structures were elucidated by spectral methods.
