Activation of the JAK/STAT pathway in Behcet’s disease [PDF]
Th1/Th17-type T-cell responses are upregulated in Behcet's disease (BD). However, signaling pathways associated with this aberrant immune response are not clarified. Whole-genome microarray profiling was performed with human U133 (Plus 2.0) chips using messenger RNA of isolated CD14(+) monocytes and CD4(+) T cells from peripheral blood mononucleated ...

Engineering IL‐4 resistant proinflammatory human myeloid cells for cancer immunotherapy
We developed a scalable workflow to generate proinflammatory human myeloid cells. CRISPR/Cas9‐edited CD34+ hematopoietic stem and progenitor cells were expanded and differentiated with M‐CSF. Deletion of STAT6 or STAT6/NFKB1 enhanced macrophage proinflammatory gene expression and cytokine secretion in the presence of IL‐4 while maintaining antibody ...

Transcripts enriched in codons that trigger P‐site tRNA‐mediated mRNA decay possess stable mRNA
PTMD codons were first described by Mendel et al. as mediators of an mRNA decay pathway dependent on the human protein CNOT3, homologous to yeast Not5. Our findings confirm that PTMD codons destabilize transcripts; however, unlike in yeast, the human pathway specifically targets and slightly destabilizes primarily stable mRNAs.

JAK-STAT signaling pathway in cancer: from molecular mechanisms to clinical intervention
The Janus kinase/signal transducer and activator of transcription (JAK/STAT) signaling pathway is a conserved signaling network that mediates communication between extracellular cytokine stimulation and nuclear transcriptional responses.

IGF2 knockout reduces but does not abolish osteosarcoma growth in vitro and in vivo
To test whether endogenous IGF2 promotes osteosarcoma growth, IGF2 was knocked out in Saos2 cells via CRISPR‐Cas9. KO cells showed reduced proliferation in vitro, and knockout xenografts in mice reached only ~25% of wild‐type tumor volume. Insulin‐like growth factor 2 (IGF2) is implicated in osteosarcoma, but direct functional evidence of its role is ...

Type I interferons modulate autophagy to shape gemcitabine response in pancreatic cancer cells
Type I interferons differentially modulate autophagy and the response of pancreatic cancer cells to gemcitabine. IFNα2b stimulates autophagic flux and protects cells from gemcitabine‐induced cell death, contributing to chemoresistance. In contrast, IFNβ1a inhibits autophagosome formation and enhances gemcitabine‐induced cell death, resulting in ...

Hipk is required for JAK/STAT activity during development and tumorigenesis.
Drosophila has been instrumental as a model system in studying signal transduction and revealing molecular functions in development and human diseases. A point mutation in the Drosophila Janus kinase JAK (called hop) causes constitutive activation of the

Intracerebral Hemorrhage Induces Monocyte TNF Signaling in Patients That Is Suppressed by BAF312
ABSTRACT Objective Intracerebral hemorrhage (ICH) causes high morbidity and mortality, with neurotoxic inflammation driven by infiltrating monocytes. This study is an in‐depth longitudinal examination of the immune response during the first week of ICH in the presence and absence of the immunomodulatory drug BAF312 (Siponimod).

Targeting the Jak/STAT pathway for immunosuppression [PDF]
While many effective immunosuppressive drugs exist and are in wide use, most target ubiquitously expressed molecules. Examples include steroids, ciclosporin A, tacrolimus, mycophenolate mofetil, and sirolimus. Consequently, these drugs have adverse effects unrelated to their immunosuppressive actions, and much effort has been directed towards ...
