Naphthyl ketones: A new class of Janus kinase 3 inhibitors
Potent inhibition of Janus kinase 3 was found for a series of naphthyl(β-aminoethyl)ketones (e.g. 7, pIC50 = 7.1 ± 0.3). Further studies indicated that these compounds fragment in less than 1 h by retro-Michael reaction in the Jak3 in vitro ELISA assay ...

In this retrospective study of 289 patients with Crohn's disease undergoing a single‐incision laparoscopy‐first (SILS‐first) strategy, conversion to open surgery was required in only 11.8% of cases. Fistula formation, colon resection, and smoking history were identified as independent risk factors for conversion, while postoperative complications and ...

Novel Role of Janus Kinase 1 in the Regulation of Oncostatin M Receptor Surface Expression [PDF]
The oncostatin M receptor (OSMR) is part of a heterodimeric receptor complex that mediates signal transduction of the pleiotropic cytokine OSM via a signaling pathway involving Janus kinases (Jaks) and transcription factors of the signal transducers and activators of transcription (STAT) family.

ABSTRACT Hypomethylating agents (HMA) and allogeneic hematopoietic stem cell transplantation (alloHSCT) have both demonstrated remissions in VEXAS; however, comparative data is lacking. We conducted a multicenter, retrospective analysis of 66 patients diagnosed with VEXAS syndrome treated with HMA (n = 35) or alloHSCT (n = 31). Baseline characteristics

From Interferon Signature to the Clinical Landscape: Type I Interferonopathies
Background Type I interferonopathies are heterogeneous diseases driven by dysregulated IFN‐I signaling. Diagnosis is challenging due to clinical/molecular variability and the need for IFN‐I quantification. Objective To characterize the clinical, immunological, genetic, molecular profiles of patients with suspected enhanced IFN‐I signaling, and assess ...

Efficacy, safety and cost‐effectiveness of CAR‐T therapy
CAR T‐cells demonstrate high efficacy in blood cancers, including ALL, MM and DLBCL. Innovations target solid tumours despite challenges such as antigen escape. Combination therapies enhance the delivery and infiltration of CAR T cells. Toxicity, cost and resistance remain major barriers to clinical use.
