Drosophila as a model system to study nonautonomous mechanisms affecting tumour growth and cell death [PDF]

open access: yes, 2018
The study of cancer has represented a central focus in medical research for over a century. The great complexity and constant evolution of the pathology require the use of multiple research model systems and interdisciplinary approaches.

Bifidobacterium Pseudolongum‐Derived Acetate Attenuates Acute Pancreatitis Through GPR43‐Mediated Suppression of M1 Macrophage Polarization

open access: yesAdvanced Science, EarlyView.
The severity of acute pancreatitis (AP) is linked to reduced abundance of Bifidobacterium pseudolongum (B. pseudolongum). Administration of B. pseudolongum or its key metabolite acetate alleviates pancreatic and systemic inflammation by suppressing M1 macrophage polarization.

Cop1 constitutively regulates c-Jun protein stability and functions as a tumor suppressor in mice [PDF]

open access: yes, 2011
Biochemical studies have suggested conflicting roles for the E3 ubiquitin ligase constitutive photomorphogenesis protein 1 (Cop 1; also known as Rfwd2) in tumorigenesis, providing evidence for both the oncoprotein c-Jun and the tumor suppressor p53 as ...

Intracellular Aβ42 Sequestration by a Serine Protease Mitigates Neurotoxicity in a Drosophila Alzheimer's Disease Model

open access: yesAdvanced Science, EarlyView.
Emerging evidence suggests that intraneuronal Aβ accumulation represents an early pathogenic event in Alzheimer's disease (AD). Using Drosophila AD model, this study shows that a nonsecreted serine protease Yip7 physically interacts with Aβ. This causes intraneuronal Aβ accumulation but surprisingly reduces the associated neurotoxicity, arguing that ...

Spatiotemporal Organization of PTK7 Diffusion on Cell Surface Facilitates Tumor Invasion and Migration

open access: yesAdvanced Science, EarlyView.
The spatiotemporal dynamics of protein kinase diffusion govern cellular signaling and functions. Although vital for active receptors, the role of pseudokinase dynamics is unclear. Here, we use single‐molecule tracking to link faster diffusion of pseudokinase PTK7 to increased tumor cell invasion and migration.

miR‐135a‐5p Is a Promising Target to Prevent the Glomerulosclerosis Associated with Podocyte Developmental Toxicity in Offspring Induced by Prenatal Dexamethasone Exposure

open access: yesAdvanced Science, EarlyView.
Prenatal dexamethasone exposure (PDE) programs persistent podocyte developmental injury and adult glomerulosclerosis. Mechanistically, glucocorticoid receptor (GR) binds the miR‐135a‐5p promoter and recruits the histone acetyltransferase p300, increasing promoter histone acetylation and sustaining miR‐135a‐5p expression. Elevated miR‐135a‐5p suppresses

JNK Regulation of Depression and Anxiety

open access: yesBrain Plasticity, 2017
Depression and anxiety are the most common mood disorders affecting 300 million sufferers worldwide. Maladaptive changes in the neuroendocrine stress response is cited as the most common underlying cause, though how the circuits underlying this response are controlled at the molecular level, remains largely unknown. Approximately 40% of patients do not

ZBTB21 Is a Dual Suppressor of Pyroptosis and MHC‐I Antigen Presentation That Promotes Tumor Immune Evasion

open access: yesAdvanced Science, EarlyView.
ZBTB21 is a transcription factor that epigenetically suppresses pyroptosis and MHC‐I antigen presentation, enabling tumor immune evasion. Genetic ablation of ZBTB21 activates pyroptotic cell death and enhances antigen presentation, recruiting CD8+ T cells to overcome immune checkpoint blockade resistance.
