Keratan sulfate inhibits its release in rabbit chondrocyte
Connective Tissue Research, 1993Keratan sulfate (KS) is one of the major glycosaminoglycans in cartilage matrix proteoglycan. We find that exogenously added KS totally blocks keratanase sensitive glycosaminoglycan release measured by radiolabeled ligand. We also find that KS release is potentially inhibited by its own presence as measured by ELISA. When KS is digested with keratanase

Monoclonal antibodies to rabbit corneal keratan sulfate proteoglycan
Current Eye Research, 1982Six mouse hybrid cell lines were isolated which secrete antibodies to rabbit corneal proteoglycan. All six antibodies interacted with the same fraction of the proteoglycan, precipitating approximately 50% of proteoglycan labelled in the protein moiety. A radioimmunoassay using these antibodies measured concentrations as low as 1 g/ml unlabelled rabbit ...

Spectroscopic Characterisation of Disaccharides Derived from Keratan Sulfates
European Journal of Biochemistry, 1995Skeletal keratan sulfates have been degraded by three independent techniques and the resultant, borohydride-reduced, disaccharides have been characterised by NMR spectroscopy. The 1H and 13C (where available) chemical shifts are reported for the following substances, where GalNAc-ol represents N-acetyl-galactosaminitol, GlcNAc-ol represents N-acetyl ...

Identification and quantitation of keratan sulfate in urine
Clinica Chimica Acta, 1974Abstract The purpose of the present artical is to indicate reliable and specific methods for identification and quantitation of keratan sulfate in urine. Thinlayer chromatography on silica gel or cellulose allows rapid and specific characterization, whereas prior isolation on Dowex 1×2 with selective elution with 3.0 M NaCl only allows quantitative ...

Impaired degradation of keratan sulfate in GM1-gangliosidosis
Clinica Chimica Acta, 1982Abstract We have prepared a new radiolabeled substrate (galactose-N-acetylglucosamine 6-sulfate-[1-3H]galactitol), from shark cartilage keratan sulfate, for an assay of acid β-galactosidase activity. Using this substrate, we found that there was a striking deficiency of β-galactosidase activity in the cultured skin fibroblasts of patients with GM1 ...

Structure and biological functions of keratan sulfate proteoglycans
1994The skeletal and corneal keratan sulfate proteoglycans show a different metabolic and structural heterogeneity. The domain structure of the carbohydrate chain has been shown to be different in various animal species. There are two major types of skeletal keratan sulfate proteoglycans with and without fucose.

Variability of keratan sulfate isolated from beef cornea
Biochimica et Biophysica Acta (BBA) - Specialized Section on Mucoproteins and Mucopolysaccharides, 1964Glycosaminoglycans were isolated from beef cornea and further fractionated on the basis of ethanol solubility. Variability of keratan sulfate was found. The forms differ in reducing activity, sulfur content, infrared spectra and specific optical rotation. Keratan sulfotransferase (3′-phosphoadenylylsulfate: keratan sulfotransferase) activity is low but

Keratan sulfate: structure, biosynthesis, and function.
Glycobiology, 2000The last 5 years have seen a marked increase in research on keratan sulfate (KS) and a concomitant increase in our understanding of the range of molecules that carry this adaptable polysaccharide. More than 15 KS-linked proteins have been identified and many of the genes encoding these have been cloned.
