IGFBP-3 inhibits Wnt signaling in metastatic melanoma cells. [PDF]

open access: yes, 2016
In previous works, we have shown that insulin-like growth factor-binding protein-3 (IGFBP-3), a tissue and circulating protein able to bind to IGFs, decreases drastically in the blood serum of patients with diffuse metastatic melanoma.

Direct inhibition of GSK3beta by the phosphorylated cytoplasmic domain of LRP6 in Wnt/beta-catenin signaling. [PDF]

open access: yesPLoS ONE, 2008
Wnt/beta-catenin signaling plays a central role in development and is also involved in a diverse array of diseases. Binding of Wnts to the coreceptors Frizzled and LRP6/5 leads to phosphorylation of PPPSPxS motifs in the LRP6/5 intracellular region and ...

Synaptic Wnt/GSK3β Signaling Hub in Autism [PDF]

open access: yes, 2016
Indexación: ScopusHundreds of genes have been associated with autism spectrum disorders (ASDs) and the interaction of weak and de novo variants derive from distinct autistic phenotypes thus making up the "spectrum." The convergence of these variants in ...

LRP5 and LRP6 are not required for protective antigen-mediated internalization or lethality of anthrax lethal toxin. [PDF]

open access: yesPLoS Pathogens, 2007
Anthrax toxin (AnTx) plays a key role in the pathogenesis of anthrax. AnTx is composed of three proteins: protective antigen (PA), edema factor, and lethal factor (LF). PA is not toxic but serves to bind cells and translocate the toxic edema factor or LF

Dickkopf-related protein 1 (Dkk1) regulates the accumulation and function of myeloid derived suppressor cells in cancer [PDF]

open access: yes, 2016
Tumor–stroma interactions contribute to tumorigenesis. Tumor cells can educate the stroma at primary and distant sites to facilitate the recruitment of heterogeneous populations of immature myeloid cells, known as myeloid-derived suppressor cells (MDSCs).

CD44 regulates Wnt signaling at the level of LRP6 [PDF]

open access: yesMolecular & Cellular Oncology, 2015
CD44 was recently identified as a positive feedback regulator of Wnt/β-catenin signaling. This regulation occurs at the level of low-density lipoprotein receptor-related protein 6 phosphorylation and membrane targeting. These findings broaden our understanding of the Wnt pathway activation process and open new perspectives for anti-CD44 therapies in ...

Wnt Signaling Downregulation Mediates T Cell Apoptosis Following OKT3-Induced T Cell Activation in Preclinical Models. [PDF]

open access: yesFASEB J
In OKT3‐treated human PBMCs, Wnt signaling was downregulated, and co‐treatment with the Wnt activator CHIR99021 reduced T cell activation and apoptosis. In humanized mice, transcriptomic profiling revealed upregulation of genes associated with T cell activation, cytokine and ROS production, and apoptosis, with downregulation of canonical Wnt signaling.

Ubiquitin-dependent folding of the Wnt signaling coreceptor LRP6 [PDF]

open access: yeseLife, 2016
Many membrane proteins fold inefficiently and require the help of enzymes and chaperones. Here we reveal a novel folding assistance system that operates on membrane proteins from the cytosolic side of the endoplasmic reticulum (ER). We show that folding of the Wnt signaling coreceptor LRP6 is promoted by ubiquitination of a specific lysine, retaining ...

Clathrin and AP2 are required for PtdIns(4,5)P2-mediated formation of LRP6 signalosomes [PDF]

open access: yes, 2014
Canonical Wnt signaling is initiated by the binding of Wnt proteins to their receptors, low-density lipoprotein-related protein 5 and 6 (LRP5/6) and frizzled proteins, leading to phosphatidylinositol (4,5)bisphosphate (PtdIns(4,5)P2) production ...

LRP6 as a biomarker of poor prognosis of breast cancer [PDF]

open access: yesGland Surgery, 2021
Recently, low-density lipoprotein receptor (LDLR)-related protein 6 (LRP6) has been the focus of molecular targeted therapy for breast cancer; however, its role in breast cancer is still controversial. The purpose of this study was to investigate the effect of LRP6 overexpression on the prognosis of breast cancer. We used immunohistochemistry to detect ...
