β‐Catenin/c‐Myc Axis Modulates Autophagy Response to Different Ammonia Concentrations

open access: yesAdvanced Biology, Volume 9, Issue 3, March 2025.
Ammonia, detoxified by the liver into urea and glutamine, impacts autophagy differently at varying levels. Low ammonia activates autophagy via c‐Myc and β‐catenin, while high levels suppress it. Using Huh7 cells and Spf‐ash mice, c‐Myc's role in cytoprotective autophagy is revealed, offering insights into hyperammonemia and potential therapeutic ...

Intrinsic Photoactive Star ZnPc–Poly(glutamate) Nanoplatforms for Multimodal Glioblastoma Therapy and Brain‐Targeted Delivery

open access: yesAdvanced Functional Materials, EarlyView.
An intrinsic photoactive star‐shaped zinc phtalocyanine‐poly(L‐glutamic acid) (ZnPc‐PGA) nanoplatform for multimodal glioblastoma (GBM) therapy and brain‐targeted elivery. A ZnPc‐PGA‐based multifunctional theranostic nanocarrier platform enables image‐guided, multimodal GBM therapy. ZnPc‐PGA nanocarriers support the integration of fluorescence imaging,

Altered levels and distribution of IGF-II/M6P receptor and lysosomal enzymes in mutant APP and APP + PS1 transgenic mouse brains [PDF]

open access: yes, 2009
The insulin-like growth factor-II/mannose-6-phosphate (IGF-II/M6P) receptor participates in the trafficking of lysosomal enzymes from the trans-Golgi network or the cell surface to lysosomes.

Functional Blood‐Brain Barrier Crossing by Biomimetic M13 Phage Vectors for Targeted Neuronal Delivery

open access: yesAdvanced Healthcare Materials, EarlyView.
This study investigates the M13 bacteriophage as a biomimetic nanovector capable of crossing in vitro models of the blood–brain barrier. By exploiting peculiar transcellular pathways, M13 avoids lysosomal degradation and preserves its structural integrity and functionality.

Structural insights into mechanisms underlying stabilization of a lysosomal enzyme through protein engineering [PDF]

open access: yesStructural Dynamics
Enzyme replacement therapies (ERTs) have been approved as treatments for several lysosomal storage disorders, which are characterized by deficiency of a single lysosomal enzyme leading to progressive substrate accumulation, perturbing lysosomal function ...

Gardiquimod Nanoemulsion Targets Cutaneous Leishmaniasis Lesions Reducing Systemic Toxicity and Parasite Burden

open access: yesAdvanced Healthcare Materials, EarlyView.
Nanoemulsion delivery of a TLR7 agonist enhances safety and promotes colocalization with Leishmania‐infected macrophages in skin lesions, enabling a ∼2‐log reduction in parasite burden. However, tissue‐level constraints and TLR‐driven regulatory feedback generate a mixed immune response that limits complete parasite clearance.

Alterations in lysosomal enzymes of the proximal tubule in gentamicin nephrotoxicity [PDF]

open access: yes, 1991
Alterations in lysosomal enzymes of the proximal tubule in gentamicin nephrotoxicity. Gentamicin accumulates in proximal tubule lysosomes, increases their number, and changes their structure.

Thematic review series: Lipid Posttranslational Modifications. Lysosomal metabolism of lipid-modified proteins

open access: yesJournal of Lipid Research, 2006
Much is now understood concerning the synthesis of prenylated and palmitoylated proteins, but what is known of their metabolic fate? This review details metabolic pathways for the lysosomal degradation of S-fatty acylated and prenylated proteins. Central

Structural basis for recognition of phosphodiester-containing lysosomal enzymes by the cation-independent mannose 6-phosphate receptor [PDF]

open access: yes, 2010
Mannose 6-phosphate (Man-6-P)-dependent trafficking is vital for normal development. The biogenesis of lysosomes, a major cellular site of protein, carbohydrate, and lipid catabolism, depends on the 300-kDa cation-independent Man-6-P ...
