Small molecule inhibitors of Mdm2 E3 ubiquitin ligase activity [PDF]

open access: yes
Half of cancers retain wild type p53 but have alterations in the pathways involved in p53 regulation. Murine double minute 2 (Mdm2) regulates p53 by acting as an E3 ubiquitin ligase, which tags p53 for degradation through the proteasome. A small molecule

Genetic dissection reveals distinct contributions of the eS31 N‐terminal domain to translational accuracy in Saccharomyces cerevisiae

open access: yesThe FEBS Journal, EarlyView.
The eukaryote‐specific N‐terminal domain (NTD) of eS31 uses two distinct strategies to maintain translation fidelity. During elongation, a positively charged “hotspot” fine‐tunes the selection of incoming aa‐tRNA. During termination, the entire NTD acts as a structural scaffold to ensure the correct positioning of the release factor eRF1.

Ploidy‐dependent modulation of DNA replication kinetics in Xenopus

open access: yesThe FEBS Journal, EarlyView.
Using an interspecies replication system with Xenopus frog species of different ploidy levels, we investigated how DNA replication adapts to polyploidization. While replication was slower in tetraploid than in diploid nuclei, dodecaploid nuclei unexpectedly replicated faster than tetraploid nuclei. Our findings show that increasing genome size does not

Senolytics and exercise: Dual modalities for rejuvenating muscle

open access: yesThe Journal of Physiology, EarlyView.
Abstract figure legend The role of senolytics on the heart and skeletal muscle. Senescent cell burden increases with ageing, disuse and disease. The senolytics dasatinib+quercetin (D+Q), navitoclax and fisetin, as well as exercise, eliminate senescent cells, reducing senescent cell burden and their senescence‐associated secretory phenotype (SASP ...

Single‐Cell Transcriptomic Identification of Drug‐Tolerant Persister Mechanisms in Capecitabine‐Treated Triple‐Negative Breast Cancer

open access: yesComputational and Systems Oncology, Volume 6, Issue 1, December 2026.
ABSTRACT Triple‐negative breast cancer (TNBC) is an aggressive subtype defined by the absence of estrogen receptor (ER), progesterone receptor (PR), and HER2 expression. This lack of receptors limits targeted therapies and contributes to high rates of recurrence.

Aging of Skeletal Muscle: From Molecular Mechanisms to Therapeutic Interventions

open access: yesMedComm, Volume 7, Issue 10, October 2026.
Skeletal muscle aging is driven by coordinated local breakdown across myofibers, stem and stromal cells, immune and vascular compartments, and neuromuscular control. These mechanisms promote mitochondrial dysfunction, inflammation, senescence, impaired regeneration, fibrosis, myosteatosis, denervation, and functional decline.

E3 Ubiquitin Ligases in MASH‐Associated Liver Fibrosis: Mechanisms and Therapeutic Opportunities

open access: yesLiver International, Volume 46, Issue 10, October 2026.
ABSTRACT Metabolic dysfunction‐associated steatohepatitis (MASH) is a major cause of progressive liver fibrosis and can ultimately lead to cirrhosis and hepatocellular carcinoma (HCC). MASH‐associated liver fibrosis develops through a complex interplay among lipotoxic hepatocyte injury, oxidative and endoplasmic reticulum stress, inflammatory ...
