Polymorphisms and mutations in the melanocortin-3 receptor and their relation to human obesity. [PDF]
Inactivating mutations in the melanocortin 3 receptor (Mc3r) have been described as causing obesity in mice, but the physiologic effects of MC3R mutations in humans have been less clear. Here we review the MC3R polymorphisms and mutations identified in humans, and the in vitro, murine, and human cohort studies examining their putative effects.

Cross-species studies implicate the melanocortin 3 receptor more strongly in the control of pubertal development than energy balance [PDF]
Hypothalamic neurons expressing either POMC or AGRP sense nutritional state directly and indirectly and transmit these neuropeptide signals to other brain centres through the melanocortin 3 and 4 receptors. MC4R is primarily concerned with the control of

Ring finger protein 11 inhibits melanocortin 3 and 4 receptor signaling [PDF]
Intact melanocortin signaling via the G protein-coupled receptors (GPCRs) melanocortin receptor 4 (MC4R) and melanocortin receptor 3 (MC3R) is crucial for body weight maintenance.

Melanocortin 1 Receptor: Structure, Function and Regulation
The melanocortin 1 receptor (MC1R) is a melanocytic Gs protein coupled receptor that regulates skin pigmentation, UV responses, and melanoma risk. It is a highly polymorphic gene, and loss of function correlates with a fair, UV-sensitive, and melanoma ...

Glucose Intake during the Neonatal Period Modulates the Central Melanocortin System with Breed-Specific Differences [PDF]
Glucose is the primary substrate for cellular energy production, and its blood concentration is tightly regulated. This study investigated the effect of glucose supplementation during the first three days post-hatching on growth performance and ...

Evaluation of hypothalamic murine and human melanocortin 3 receptor transcript structure. [PDF]
The melanocortin 3 receptor (MC3R) is involved in regulation of energy homeostasis. However, its transcript structure is not well understood. We therefore studied initiation and termination sites for hypothalamic murine Mc3r and human MC3R transcripts.

The melanocortin receptors as targets for general obesity: contextualizing clinical failures and analyzing future perspectives [PDF]
The melanocortin system is a genetically conserved and clinically validated target for the treatment of obesity. Hundreds of studies establish the role of the melanocortin receptors, particularly the melanocortin-4 receptor, in food intake, weight ...

Functions of DPLIY motif and helix 8 of human melanocortin-3 receptor. [PDF]
The melanocortin-3 receptor (MC3R) is a member of the family A G protein-coupled receptors (GPCRs). The MC3R remains the most enigmatic of the melanocortin receptors with regard to its physiological functions, especially its role in energy homeostasis.

Dual activation of MC3R and MC4R drives weight loss and reduces food intake in male primates with obesity [PDF]
The melanocortin system plays a central role in regulating hunger and satiety, making it an attractive target for treating metabolic disease. However, the limited clinical success of selective melanocortin-4 receptor (MC4R) agonists has prompted ...

Melanocortin‐3 receptors are involved in adaptation to restricted feeding [PDF]
The central nervous melanocortin system forms a neural network that maintains energy homeostasis. Actions involving neural melanocortin‐3 receptors (MC3Rs) regulate the expression rhythms in ingestive behaviors and metabolism anticipating nutrient intake.
