Peroxidasin enables melanoma immune escape by inhibiting natural killer cell cytotoxicity

open access: yesMolecular Oncology, EarlyView.
Peroxidasin (PXDN) is secreted by melanoma cells and binds the NK cell receptor NKG2D, thereby suppressing NK cell activation and cytotoxicity. PXDN depletion restores NKG2D signaling and enables effective NK cell–mediated melanoma killing. These findings identify PXDN as a previously unrecognized immune evasion factor and a potential target to improve

36 | Neoadjuvant strategy for locally advanced conjunctival melanoma: a single-center approach to eye preservation

open access: yesDermatology Reports
Background: Conjunctival melanoma is a rare but aggressive ocular tumor with high metastatic potential leading to 30% mortality rate. Standard treatment relies on surgery and brachytherapy, but local recurrence and mutilating procedures (e.g ...

Transcriptional downregulation of MHC class I and melanoma de- differentiation in resistance to PD-1 inhibition

open access: yesNature Communications, 2020
Transcriptomic signatures designed to predict melanoma patient responses to PD-1 blockade have been reported but rarely validated. We now show that intra-patient heterogeneity of tumor responses to PD-1 inhibition limit the predictive performance of ...

Biological therapy in the treatment of melanoma [PDF]

open access: yes, 2018
Melanoma is one of the most aggressive tumors and its incidence is on the rise. The low rates of survival in metastatic melanoma has led to the development of new drugs for this type of patient, such as biological therapy which has shown remarkable ...

Crucial parameters for precise copy number variation detection in formalin‐fixed paraffin‐embedded solid cancer samples

open access: yesMolecular Oncology, EarlyView.
This study shows that copy number variations (CNVs) can be reliably detected in formalin‐fixed paraffin‐embedded (FFPE) solid cancer samples using ultra‐low‐pass whole‐genome sequencing, provided that key (pre)‐analytical parameters are optimized.

17 | The role of sentinel lymph node biopsy in pt1a, pt1b and pt2a melanomas: a 10-year experience through 2025

open access: yesDermatology Reports
Background: Sentinel lymph node biopsy (SLNB) is key for melanoma staging, but its value in thinner lesions is debated, with current predictive models often underestimating positivity. Methods: We retrospectively analyzed 511 patients with pT1a (8.6%),

Tumor mutational burden as a determinant of metastatic dissemination patterns

open access: yesMolecular Oncology, EarlyView.
This study performed a comprehensive analysis of genomic data to elucidate whether metastasis in certain organs share genetic characteristics regardless of cancer type. No robust mutational patterns were identified across different metastatic locations and cancer types.

03 | Pancreatic cancer burden in Italian CDKN2a-positive families followed for up to 25 years

open access: yesDermatology Reports
Background: Pancreatic cancer (PC) risk in CDKN2A pathogenic variant (PV) carriers is among the highest (more than twice that of BRCA1/2), with a standardized incidence ratio (SIR) of 20 and cumulative incidence of 20% by age 70 in the Dutch cohort ...

The PI3Kδ inhibitor roginolisib (IOA‐244) preserves T‐cell function and activity

open access: yesMolecular Oncology, EarlyView.
Identification of novel PI3K inhibitors with limited immune‐related adverse effects is highly sought after. We found that roginolisib and idelalisib inhibit chronic lymphocytic leukemia (CLL) cells and Treg suppressive functions to similar extents, but roginolisib affects cytotoxic T‐cell function and promotion of pro‐inflammatory T helper subsets to a

Targeting p38α in cancer: challenges, opportunities, and emerging strategies

open access: yesMolecular Oncology, EarlyView.
p38α normally regulates cellular stress responses and homeostasis and suppresses malignant transformation. In cancer, however, p38α is co‐opted to drive context‐dependent proliferation and dissemination. p38α also supports key functions in cells of the tumor microenvironment, including fibroblasts, myeloid cells, and T lymphocytes.
