Neuropathic Pain: From Mechanisms to Therapeutic Interventions

open access: yesMedComm, Volume 7, Issue 10, October 2026.
Neuropathic pain arises from peripheral nerve injury, central sensitization, neurotransmitter imbalance, neuroimmune activation, and maladaptive circuit remodeling. Clinical phenotyping and multimodal imaging can identify dominant peripheral and central mechanisms.

Brain Penetrant Calcium Channel Blockers Were Not Associated With Reduced Alcohol Consumption: Converging Results From Two Large Independent Cohort Studies Using Electronic Health Records

open access: yesPharmacoepidemiology and Drug Safety, Volume 35, Issue 10, October 2026.
ABSTRACT Purpose Alcohol use disorder (AUD) remains a major public health problem, with few effective treatments. L‐type calcium channel blockers (LTCCBs) have genetic and preclinical support as potential treatments for AUD. Methods We evaluated whether brain penetrant (BP)‐LTCCBs are associated with reduced alcohol consumption in two preregistered ...

Lasting reduction in mesolimbic dopamine neuronal activity after morphine withdrawal

open access: yes, 1999
The activity of mesolimbic dopaminergic neurons was investigated in rats at various times after a chronic regimen of morphine, which produced, upon suspension, a marked somatic withdrawal syndrome.

Dualsteric and dual‐acting modulation of muscarinic receptors by antagonist KH‐5

open access: yesBritish Journal of Pharmacology, Volume 183, Issue 19, Page 5769-5790, October 2026.
Abstract Background and purpose Muscarinic acetylcholine receptors are key therapeutic targets, and ligands engaging both orthosteric and allosteric sites may offer improved selectivity and efficacy. Here, we investigated whether the muscarinic antagonist KH‐5 acts as a dualsteric antagonist and defined its mode of interaction with muscarinic receptors.

Muscarinic agonists for schizophrenia: Bridging preclinical evidence and clinical enthusiasm

open access: yesBritish Journal of Pharmacology, Volume 183, Issue 19, Page 5701-5731, October 2026.
Abstract Antipsychotic drugs targeting dopamine receptors have been used to treat schizophrenia for decades, primarily alleviating positive symptoms such as hallucinations and delusions. However, they provide limited benefit for cognitive and negative symptoms—major contributors to poor outcomes—and are frequently associated with significant side ...

Intermittent cocaine use patterns, not total intake, predict cue‐induced drug seeking in rats

open access: yesBritish Journal of Pharmacology, Volume 183, Issue 20, Page 6094-6112, October 2026.
Abstract Background and Purpose Cocaine‐associated cues trigger relapse to drug use in humans and animal models. In rats, long daily cocaine access (4–6 vs. 1–2 h) increases drug self‐administration and cue‐induced cocaine seeking, suggesting that greater intake promotes relapse.

Mesolimbic dopaminergic decline after cannabinoid withdrawal [PDF]

open access: yes, 1998
The mesolimbic dopamine system has recently been implicated in the long-term aversive consequences of withdrawal from major drugs of abuse. In the present study we sought to determine whether mesolimbic dopamine neurons are involved in the ...

Early Weight Regain After GLP‐1 Receptor Agonist Discontinuation: Mechanisms and Implications for Treatment De‐Escalation Strategies

open access: yesDiabetes, Obesity and Metabolism, Volume 28, Issue 10, Page 8769-8776, October 2026.
ABSTRACT Glucagon‐like peptide‐1 receptor agonists (GLP‐1RAs) have transformed the pharmacological management of obesity, producing substantial and sustained weight loss during active treatment. However, discontinuation of therapy is frequently followed by significant weight regain, raising important questions regarding the durability of treatment ...

The Role of Kappa‐Opioid Receptor/Dynorphin System in Modulating Neural‐Circuits of Depression and Anxiety—A Review

open access: yesJournal of Cellular and Molecular Medicine, Volume 30, Issue 19, October 2026.
ABSTRACT Major depression and anxiety disorders are psychiatric disorders that involve a great number of people. There is a growing need for new and better pharmacological treatments for these diseases. Kappa‐opioid receptor (KOR) antagonists seem to be a promising new pathway for the development of new antidepressants and anxiolytics, but a great ...

Acute ayahuasca administration attenuates binge‐like eating and anxiety‐related behaviors in mice subjected to intermittent fasting

open access: yesJournal of Neuroendocrinology, Volume 38, Issue 10, October 2026.
Abstract Human binge eating disorder is characterized by recurrent episodes of excessive food consumption accompanied by loss of control and is strongly associated with alterations in reward processing and emotional regulation. Ayahuasca, a psychoactive botanical preparation traditionally used in Amazonian medicine, has been shown to modulate ...
