Inhibition of mTORC1 through ATF4-induced REDD1 and Sestrin2 expression by Metformin

open access: yesBMC Cancer, 2021
Background Although the major anticancer effect of metformin involves AMPK-dependent or AMPK-independent mTORC1 inhibition, the mechanisms of action are still not fully understood.

Advances in Preventive Therapy for Estrogen-Receptor-Negative Breast Cancer. [PDF]

open access: yes, 2014
Preventing breast cancer is an effective strategy for reducing breast cancer deaths. The purpose of chemoprevention (also termed preventive therapy) is to reduce cancer incidence by use of natural, synthetic, or biological agents.

Alkyltriphenylphosphonium Binding to Cardiolipin Triggers Oncosis in Cancer Cells

open access: yesAdvanced Science, EarlyView.
Alkyltriphenylphosphonium, exemplified by TPP+‐C14, preferentially accumulates in mitochondria and selectively binds to cardiolipin, a key phospholipid of the inner mitochondrial membrane, causing loss of mitochondrial membrane potential, severe cellular ATP depletion, and calcium imbalance.

Valproic acid sensitizes metformin-resistant human renal cell carcinoma cells by upregulating H3 acetylation and EMT reversal

open access: yesBMC Cancer, 2018
Background Metformin (Met) is a widely available diabetic drug and shows suppressed effects on renal cell carcinoma (RCC) metabolism and proliferation. Laboratory studies in RCC suggested that metformin has remarkable antitumor activities and seems to be

Metformin Restores Tetracyclines Susceptibility against Multidrug Resistant Bacteria

open access: yesAdvancement of science, 2020
Highly persistent incidence of multidrug resistant (MDR) bacterial pathogens constitutes a global burden for public health. An alternative strategy to alleviate such a crisis is to identify promising compounds to restore antibiotics activity against MDR ...

Dipiperazine‐Phenyl Derivatives Based on Convergent Molecular Platforms Can Reverse Multidrug Resistance in Gram‐Negative Bacteria by Inhibiting Efflux and Permeabilizing Cell Membranes

open access: yesAdvanced Science, EarlyView.
By integrating a convergent molecular platform strategy, this study designed a novel dual‐target C5 to combat multidrug‐resistant Gram‐negative bacteria. C5 synergistically enhances antibiotic efficacy by inhibiting efflux pumps and increasing bacterial membrane permeability.

Disrupting the Formation of YAP Condensates Promotes the Activation of AMPKα to Inhibit the Progression of Primary Liver Cancer

open access: yesAdvanced Science, EarlyView.
This study systematically investigates the function and molecular mechanisms of YAP phase separation in multiple primary liver cancers. These findings provide novel insights into phase separation‐mediated primary liver cancer development and validate targeted disruption of this process as an effective therapeutic strategy for primary liver cancer ...

Metformin reverses established lung fibrosis in a bleomycin model

open access: yesNature Medicine, 2018
Fibrosis is a pathological result of a dysfunctional repair response to tissue injury and occurs in a number of organs, including the lungs1. Cellular metabolism regulates tissue repair and remodelling responses to injury2–4. AMPK is a critical sensor of

Targeting Lactate and Lactylation in Cancer Metabolism and Immunotherapy

open access: yesAdvanced Science, EarlyView.
Lactate, once deemed a metabolic waste, emerges as a central regulator of cancer progression. This review elucidates how lactate and its epigenetic derivative, protein lactylation, orchestrate tumor metabolism, immune suppression, and therapeutic resistance.

Impact of metformin, statin, aspirin and insulin on the prognosis of uHCC patients receiving first line Lenvatinib or Atezolizumab plus Bevacizumab

open access: yesScientific Reports
Recently, in Hepatocellular carcinoma (HCC) setting, the use of metformin has been associated to a trend toward worse response rate, overall survival and progression free survival in patients who received immunotherapy.
