In Parkinson's disease, SHMT1 downregulation disrupts its interaction with PEMT in astrocytes, reducing SAM levels. This leads to H3K4me1 hypomethylation and decreased Slc1a2/Glul expression, ultimately exacerbating neuroexcitotoxicity and dopaminergic neuron loss.

Objective Gout caused by the formation of monosodium urate (MSU) crystals and calcium pyrophosphate (CPP) deposition disease are two major types of microcrystalline pathologies in adults. They are responsible for recurrent flares that rely on interleukin (IL) 1β via activation of the NLRP3 inflammasome.

Consequences of the divergence of Methionine AdenosylTransferase [PDF]
AbstractMethionine adenosyltransferase (MAT), which catalyzes the biosynthesis of S-adenosylmethionine from L-methionine and ATP, is an ancient, highly conserved enzyme present in all three domains of life. Although the MAT enzymes of each domain are believed to share a common ancestor, the sequences of archaeal MATs show a high degree of divergence ...

This study identifies SPTAN1‐kla as a novel oncogenic driver in HBV‐driven HCC, linking metabolic, epigenetic, and immune mechanisms. SPTAN1‐kla promotes HCC progression via NOTCH1/HES1 activation and immunosuppression. Findings highlight that targeting SPTAN1‐kla provides a framework for HCC treatment to improve clinical outcomes in HBV infected ...

USP30 deubiquitinates and stabilizes MAT2A, thereby regulating DNA methylation and miRNA expression, which ultimately leads to disruption of lung endothelial barrier and inflammation. Abstract Microvascular dysfunction is a key contributor to the development of acute inflammatory diseases, characterized by heightened vascular hyperpermeability and ...

A simplified chemoenzymatic platform is designed to integrate an enzyme cascade with subsequent chemical steps, enabling the divergent production of diverse benzylisoquinoline alkaloids (BIAs). Demonstrating remarkable versatility and enantioselectivity, four distinct classes of BIAs are efficiently generated, in which the natural medicine papaverine ...

Methionine Adenosyltransferase Purified from Rat Liver
Methionine adenosyltransferase (MAT III), also known as S-adeno-sylmethionine synthetase, was purified from rat liver and crystallized. X-ray diffraction data were collected using a microfocused synchrotron radiation. The crystallization conditions were extensively optimized but final crystal size was never larger than 303 pm3.
