A human H5N1 influenza virus expressing bioluminescence for evaluating viral infection and identifying therapeutic interventions

open access: yesiScience
Summary: A multistate outbreak of highly pathogenic avian influenza virus (HPAIV) H5N1 in the United States dairy cattle was first reported on March 2024, followed by a zoonotic cattle-to-human virus transmission to a dairy farm worker in Texas.

Cell‐cycle‐specific lesion evolution rather than inhibition of double‐strand‐break repair underpins cisplatin radiosensitization

open access: yesMolecular Oncology, EarlyView.
We analyze cisplatin–DNA adducts (CDAs) and double‐strand breaks (DSBs) in a cell‐cycle‐dependent manner. We find that CDAs form similarly across all cell cycle phases. DSBs arise only in S‐phase. CDAs might not directly impair DSB repair, but S‐phase DSB lesions evolve in the presence of CDAs and disrupt repair in G2, also causing radiosensitization ...

Longitudinal circulating tumor DNA profiling in patients with advanced endometrial cancer using an off‐the‐shelf targeted NGS panel

open access: yesMolecular Oncology, EarlyView.
Intratumour heterogeneity complicates precision management of advanced endometrial cancer. Circulating tumor DNA (ctDNA) offers a minimally invasive strategy to capture tumor evolution and therapeutic resistance. Here, we compare tumor‐agnostic NGS with tumor‐informed ddPCR, outlining their relative sensitivity, concordance, and clinical implications ...

CRISPR/dCas9-targeted H3K27me3 demethylation at the CUC3 boundary gene triggers ectopic transcription and impacts plant development

open access: yesiScience
Summary: Chromatin modifications are deemed to correlate with gene expression patterns, yet their direct causal effect on transcription and cell fate remains unestablished. The H3K27me3 modification, highly conserved in eukaryotes, is strongly associated

Interaction of HS1BP3 with cortactin modulates TKS5 localisation, cell secretion and cancer malignancy

open access: yesMolecular Oncology, EarlyView.
Here, we demonstrate that HS1BP3 interacts with Cortactin through a proline‐rich region (PRR3.1) and show that this interaction, and HS1BP3 itself, promote cancer cell proliferation and invasion. Inhibition of this interaction leads to build‐up of TKS5 in multivesicular endosomes and altered secretion of CD63 and CD9, providing an explanation for the ...

Representations and generalization in artificial and brain neural networks

open access: yesProceedings of the National Academy of Sciences of the United States of America
Humans and animals excel at generalizing from limited data, a capability yet to be fully replicated in artificial intelligence. This perspective investigates generalization in biological and artificial deep neural networks (DNNs), in both in-distribution
