Histone methyltransferases in cancer

Seminars in Cell & Developmental Biology, 2010
Cancer is perceived as a heterogeneous group of diseases that is characterized by aberrant patterns of gene expression. In the last decade, an increasing amount of data has pointed to a key role for epigenetic alterations in human cancer. In this review, we focus on a subclass of epigenetic regulators, namely histone methyltransferases (HMTs).

Immobilization of S-Methyltransferase

Enzyme, 2017
S-methyltransferase was solubilized from pig liver microsomes by treatment with N-dodecyl-N,N-dimethyl-3-ammonio-l-sulfonate (Zwittergent®). The soluble enzyme was immobilized by covalent binding to agarose and by copolymerization with acrylamide. The specific activity for the agarose-bound enzyme towards the substrate ethane thiol was 0.87 nmol/min/mg

Plant DNA methyltransferases

Plant Molecular Biology, 2000
DNA methylation is an important modification of DNA that plays a role in genome management and in regulating gene expression during development. Methylation is carried out by DNA methyltransferases which catalyse the transfer of a methyl group to bases within the DNA helix.

Elongation factor methyltransferase 3 – A novel eukaryotic lysine methyltransferase

Biochemical and Biophysical Research Communications, 2014
Here we describe the discovery of Saccharomycescerevisiae protein YJR129Cp as a new eukaryotic seven-beta-strand lysine methyltransferase. An immunoblotting screen of 21 putative methyltransferases showed a loss in the methylation of elongation factor 2 (EF2) on knockout of YJR129C.

Cobalamin-Dependent Methyltransferases

Accounts of Chemical Research, 2001
Cobalamin cofactors play critical roles in radical-catalyzed rearrangements and in methyl transfers. This Account focuses on the role of methylcobalamin and its structural homologues, the methylcorrinoids, as intermediaries in methyl transfer reactions, and particularly on the reaction catalyzed by cobalamin-dependent methionine synthase.

Lysine Methyltransferases and Their Inhibitors

2019
Since 2000, the histone methyltransferases that catalyze the methylation of a number of histone and nonhistone substrates have been discovered.

DNA modification by methyltransferases

Current Opinion in Structural Biology, 1995
Enzymatic methylation of DNA plays important roles in both prokaryotes and eukaryotes. Structural study of the HhaI DNA methyltransferase has provided considerable insight into the chemistry of C5-cytosine methylation. The DNA-protein complex reveals a substrate cytosine flipped out of the double helix during the reaction, and a novel two-loop DNA ...

The Actin Histidine methyltransferase SETD3 is a CHD1 lysine di-methyltransferase

Cancer Letters
Protein methylation is a widespread posttranslational modification that primarily targets lysine, arginine, and histidine residues. Aberrant protein methylation has been implicated in tumorigenesis, although the specific role of SETD3, a histidine methyltransferase, in cancer remains poorly understood.

REGULATION OF PHENYLETHANOLAMINE N-METHYLTRANSFERASE

Biochemical Pharmacology, 1973
Publisher Summary This chapter discusses regulation of phenylethanolamine N-methyltransferase (PNMT). PNMT is localized almost exclusively to the adrenal medulla, although traces of activity have been reported in heart, brain, in extra-adrenal chromaffin tissue and in sympathetic ganglia.
