In higher plants isoprenoids have many essential roles e.g. in membrane structure (phytosterols), redox chemistry (plastoquinone) and as antioxidants (carotenoids). Despite their functional and chemical diversity all isoprenoids are biosynthetically related from a common precursor, isopentenyl pyrophosphate (IPP).

Only recently, the existence of a second mevalonate-independent pathway for the biosynthesis of IPP and DMAPP was detected in plants and certain eubacteria.

Targeting mutant p53 through the mevalonate pathway

It is well established that mutant forms of the p53 tumour suppressor acquire pro-oncogenic activities. Inhibition of the mevalonate pathway is now shown to promote degradation of select oncogenic mutant p53 proteins, indicating that destabilization of mutant p53 could be a promising therapeutic strategy.
