The mechanism for sustained Ca2+ influx activated by G protein–coupled receptors was examined. In Chinese hamster ovary cells expressing recombinant human endothelin type B receptor (ETBR) and endogenous P2Y receptor (P2Y-R), endothelin-1 elicited a ...

The regulation of AP-1 activity by mitogen-activated protein kinases [PDF]
AP-1 is a collection of dimeric sequence specific, DNA binding, transcriptional activators composed of Jun and Fos subunits. The composition, the level and the activity of AP-1 complexes are regulated in response to extracellular stimuli. An important role in this regulation is played by mitogen-activated protein kinases (MAPKs).

Manganese modulation of MAPK pathways: effects on upstream mitogen activated protein kinase kinases and mitogen activated kinase phosphatase‐1 in microglial cells [PDF]
AbstractMultiple studies demonstrate that manganese (Mn) exposure potentiates inflammatory mediator output from activated glia; this increased output is associated with enhanced mitogen activated protein kinase (MAPK: p38, ERK and JNK) activity. We hypothesized that Mn activates MAPK by activating the kinases upstream of MAPK, i.e. MKK‐3/6, MKK‐1/2 and

Mitogen-activated Protein Kinase Phosphorylates and Regulates the HIV-1 Vif Protein [PDF]
The human immunodeficiency virus type 1 (HIV-1) Vif protein plays a critical role in virus replication and infectivity. Here we show that Vif is phosphorylated and regulated by p44/42 mitogen-activated protein kinase (MAPK). Vif phosphorylation by MAPK was demonstrated in vitro as well as in vivo and was shown to occur on serine and threonine residues.

Mitogen-Activated Protein Kinase Kinase Kinase 1 (MAP3K1) is a key signaling molecule essential for eyelid closure during embryogenesis. In mice, Map3k1 knockout leads to a fully penetrant eye-open at birth (EOB) phenotype due to disrupted MAPK signaling,

Although spleen tyrosine kinase (Syk) has crucial roles in various cells, its function on vascular smooth muscle contraction has not been determined. In the present study, we performed experiments to determine if Syk contributes to the endothelin-1 (ET-1)

Mitogen regulation of c-Raf-1 protein kinase activity toward mitogen-activated protein kinase-kinase
The c-raf-1 protooncogene encodes a Ser/Thr protein kinase. A mitogen-activated protein kinase-kinase (MAPKK) purified from bovine brain is phosphorylated and activated 4-9-fold in vitro by c-Raf-1 from mitogen-treated cells. c-Raf-1 protein kinase activity, measured by the phosphorylation of brain MAPKK substrate, is detectably activated within 1 min ...

Regulation of the mitogen-activated protein kinase kinase (MEK)-1 by NAD+-dependent deacetylases [PDF]
Sirtuins are class III deacetylases that regulate many essential processes, including cellular stress, genome stability and metabolism. Although these NAD(+)-dependent deacetylases control adaptive cellular responses, identification of sirtuin-regulated signaling targets remain under-studied.

Pervanadate inhibits mitogen‐activated protein kinase kinase‐1 in a p38MAPK‐dependent manner [PDF]
In baboon smooth muscle cells (SMCs), pervanadate has a biphasic dose‐dependent effect on MEK‐1 activity. After a 30 min incubation period, low concentrations (1–10 μM) activate, while higher doses (30–100 μM) fail to stimulate MEK‐1. One possibility is that higher doses of pervanadate induce an additional signaling pathway that inhibits MEK‐1.
