A mixed-mode chromatographic (MMC) sorbent was prepared by functionalizing the silica sorbent with a pentafluorophenyl (PFP) ligand. The resulting stationary phase provided a reversed-phase (RP) retention mode along with a relatively mild strong cation-exchange (SCX) retention interaction. While the mechanism of interaction is not entirely clear, it is

Preparation and evaluation of dextran-grafted mixed-mode chromatography adsorbents
Journal of Chromatography A, 2019In the present work, mixed-mode chromatography adsorbents grafted with dextran of different molecular weight were prepared. Compared to non-grafted adsorbent (BA-6B), the morphology of dextran-grafted adsorbents (BA-6B-Ts) did not change significantly, while the water content (ω), porosity (P) and pore volume (V) decreased.

Mixed-Mode Chromatography and Its Role in Monoclonal Antibody Purification
2023As the biopharmaceutical industry matures and embraces process intensification methodologies allied to the emergence of newer personalized medicines, a key constant is the regulatory need to purify products that satisfy the criteria of safety, quality, and efficacy in each batch of released product destined for clinical use.

Antibody Aggregate Removal Using a Mixed-Mode Chromatography Resin
2020In monoclonal antibody (mAb) production, aggregates represent a major class of product-related impurities that needs to be removed by the downstream process. Protein A chromatography is generally less effective at removing antibody aggregates under typical conditions, and in most cases aggregate removal relies on a subsequent polishing chromatography ...

Polydopamine‐functionalized monolithic silica column for mixed‐mode chromatography†
Journal of Separation Science, 2013Dopamine is easy to self‐polymerize under alkaline conditions and the resultant polydopamine is easy to adhere to the surface of many organic and inorganic materials. Based on the characteristics of dopamine, in this paper, a new polydopamine functionalized monolithic silica column was successfully prepared for performing mixed‐mode chromatography. The

Analysis of monoclonal antibody oxidation by simple mixed mode chromatography
Journal of Chromatography A, 2016Analysis of oxidation of monoclonal antibodies (mAbs) in most cases relies on peptide mapping and LC-MS, which is time consuming and labor-intensive. A robust chromatography based method that is able to resolve and quantitate mAb oxidation variants due to oxidized methionine or tryptophan is highly desired.

Purification process of recombinant monoclonal antibodies with mixed mode chromatography
Journal of Chromatography A, 2015An innovative process to purify mAb from CHO cell culture supernatant was developed. This three-step process involved two mixed mode resins and an anion exchange membrane. We used a human IgG mixture to determine the optimal conditions for each purification step.

Two-dimensional liquid chromatography with mixed mode stationary phases
Journal of Chromatography A, 2005Mixed mode stationary phases with ion-pairing reagent (acidic or basic) as integral part of hydrophobic chain offers unique selectivity, and hence, are ideal for multidimensional separations. The retention of hydrophobic components is a function of organic content, whereas that of charged species is a function of organic content, ionogenic modifier and

Mixed‐mode separation of polycyclic aromatic hydrocarbons (PAHs) in electrokinetic chromatography
ELECTROPHORESIS, 1998AbstractA mixed‐mode separation technique has been developed and optimized for the separation of the 16 Environmental Protection Agency (EPA) priority polycyclic aromatic hydrocarbons (PAHs). The procedure utilized two different buffer additives as pseudo‐stationary phases with different selectivities towards the analytes. Sodium dioctyl sulfosuccinate
