Combined physical activity in the prevention of cardiovascular diseases: from physiological to molecular adaptation mechanisms

open access: yesКлиническая практика
Physical activity is recognized as the most important non-medicinal tool for the prevention of cardiovascular diseases, however, the highest efficiency is demonstrated by the programs combining various types of physical load.

Hippo pathway at the crossroads of stemness and therapeutic resistance in breast cancer

open access: yesMolecular Oncology, EarlyView.
Dysregulation of the Hippo pathway drives nuclear accumulation of YAP/TAZ, activating stemness‐related transcriptional programs that sustain breast cancer stemness and fuel therapeutic resistance across subtypes, underscoring Hippo signaling as a targetable vulnerability. Figure created and edited with BioRender.com.

Polyphenol-rich extract from Wisteria floribunda (Willd.) DC. flowers promotes osteoblast differentiation and bone formation by modulating the mTOR pathway

open access: yesAsian Pacific Journal of Tropical Biomedicine
Objective: To investigate the osteogenic effects of polyphenol-rich extracts from Wisteria floribunda (Willd.) DC. (W. floribunda) flowers and elucidate the underlying mechanisms. Methods: Polyphenolic compounds of W.

Pathogens MenTORing Macrophages and Dendritic Cells: Manipulation of mTOR and Cellular Metabolism to Promote Immune Escape

open access: yesCells, 2020
Myeloid cells, including macrophages and dendritic cells, represent an important first line of defense against infections. Upon recognition of pathogens, these cells undergo a metabolic reprogramming that supports their activation and ability to respond ...

E2A selectively regulates TGF‐β–induced apoptosis in KRAS‐mutant non‐small cell lung cancer

open access: yesMolecular Oncology, EarlyView.
Ability to induce apoptosis by TGF‐β is frequently lost in advanced lung adenocarcinoma despite intact TGF‐β signaling. We identify E2A as a mutant KRAS–dependent mediator of resistance to TGF‐β–induced apoptosis. TGF‐β induces E2A via SMAD3 in mutant KRAS cells, and E2A silencing restores apoptosis and enhances radiation response in cell lines ...

Deconvoluting mTOR biology [PDF]

open access: yesCell Cycle, 2012
In metazoans, TOR is an essential protein that functions as a master regulator of cellular growth and proliferation. Over the past decade, there has been an explosion of information about this critical master kinase, ranging from the composition of the TOR protein complex to its ability to act as an integrator of numerous extracellular signals ...

Adaptor protein CIN85 potentiates the motility of osteosarcoma cells via the Akt/mTOR and MMP2‐COL3A1 axis

open access: yesMolecular Oncology, EarlyView.
CIN85 is highly expressed in osteosarcoma, particularly in metastatic lesions. Its overexpression increases cell migration and Matrigel invasion, while silencing CIN85 suppresses these behaviors. Transcriptome analysis shows that CIN85 regulates MMP2, COL3A1, and Akt/mTOR signaling. Targeting these pathways reverses CIN85‐induced motility, highlighting

Influence of oxidative stress on the activity of mTOR kinase in CD4+T lymphocytes in autoimmune thyroiditis

open access: yesМедицинская иммунология
Currentstudies of autoimmunityprocesses areaddressing specific processes involving mitochondria, i.e., altered intracellular signaling including the generation of ROS and release of proteins from mitochondria to the cytoplasm, thus activating apoptosis ...

Upstream and downstream of mTOR [PDF]

open access: yesGenes & Development, 2004
The evolutionarily conserved checkpoint protein kinase, TOR (target of rapamycin), has emerged as a major effector of cell growth and proliferation via the regulation of protein synthesis. Work in the last decade clearly demonstrates that TOR controls protein synthesis through a stunning number of downstream targets.
