TLR7-mediated skin inflammation remotely triggers chemokine expression and leukocyte accumulation in the brain [PDF]

open access: yes, 2016
Background: The relationship between the brain and the immune system has become increasingly topical as, although it is immune-specialised, the CNS is not free from the influences of the immune system.

Embedded CRISPRi Enhances Gene‐Silencing Efficiency in Drosophila

open access: yesAdvanced Science, EarlyView.
Current CRISPR interference (CRISPRi) technology in Drosophila has limited efficiency. This study introduces the emCRISPRi platform, which significantly enhances transcriptional silencing efficacy by embedding inhibitory domains within the dCas9 architecture.

Direct PCR with the CDC 2019 SARS-CoV-2 assay: optimization for limited-resource settings

open access: yesScientific Reports, 2022
PCR-based diagnostics generally require nucleic acid extraction from patient specimens prior to amplification. As highlighted early in the COVID-19 pandemic, extraction steps may be difficult to scale during times of massive demand and limited reagent ...

Atypical chemokine receptor ACKR2 controls branching morphogenesis in the developing mammary gland [PDF]

open access: yes, 2016
Macrophages are important regulators of branching morphogenesis during development and postnatally in the mammary gland. Regulation of macrophage dynamics during these processes can therefore have a profound impact on development.

The FOXC2‐LAMA4 Axis Orchestrates Vasculogenic Mimicry and Immunosuppressive Niche Formation to Drive Metastatic Cascade in Renal Cell Carcinoma

open access: yesAdvanced Science, EarlyView.
Advanced clear cell renal cell carcinoma metastasis is driven by a FOXC2+ tumor subpopulation. These cells execute vasculogenic mimicry for dissemination and secrete LAMA4. LAMA4 engages macrophage ITGA6, activating the STAT6/GATA3 axis to polarize macrophages into a pro‐metastatic, immunosuppressive phenotype.

Quantitative Multiplex Real-Time Reverse Transcriptase–Polymerase Chain Reaction with Fluorescent Probe Detection of Killer Immunoglobulin-Like Receptors, KIR2DL4/3DL3

open access: yesDiagnostics, 2020
(1) Background: KIR2DL4/KIR3DL3 are the framework genes present in all KIR haplotypes, with unique expression patterns being present only in women and CD56bright NK cells. KIR genes have a high degree of DNA sequence identity.

Effects of local cryotherapy on systemic endothelial activation, dysfunction, and vascular inflammation in adjuvant-induced arthritis (AIA) rats

open access: yesArthritis Research & Therapy, 2022
Aim This study explored the systemic vascular effects of local cryotherapy with a focus on endothelial changes and arterial inflammation in the model of rat adjuvant-induced arthritis (AIA). Methods Cryotherapy was applied twice a day on hind paws of AIA

Improving statistical inference on pathogen densities estimated by quantitative molecular methods: malaria gametocytaemia as a case study [PDF]

open access: yes, 2015
BACKGROUND: Quantitative molecular methods (QMMs) such as quantitative real-time polymerase chain reaction (q-PCR), reverse-transcriptase PCR (qRT-PCR) and quantitative nucleic acid sequence-based amplification (QT-NASBA) are increasingly used to ...

Oral Streptococcus salivarius Couples Neutrophil IRGM1 Signaling to NET Formation and Colorectal Cancer Metastasis

open access: yesAdvanced Science, EarlyView.
The oral bacterium Streptococcus salivarius promotes colorectal cancer metastasis by inducing neutrophil extracellular trap (NET) formation. Mechanistically, IRGM1–IQGAP1 interaction activates Wnt5a–PI3K/AKT signaling in neutrophils, driving NET‐mediated tumor progression.

Yellow fever in the diagnostics laboratory [PDF]

open access: yes, 2018
Yellow fever (YF) remains a public health issue in endemic areas despite the availability of a safe and effective vaccine. In 2015-2016, urban outbreaks of YF were declared in Angola and the Democratic Republic of Congo, and a sylvatic outbreak has been ...
