SAMHD1 regulates intracellular dNTP pools, influencing lentiviral transduction, gene therapy efficiency, and disease progression in pediatric hematological malignancies. Integrated bioinformatics and targeted strategies, including CRISPR and pharmacological inhibition, highlight its therapeutic potential.

ABSTRACT Cancer remains a major cause of global mortality, necessitating the identification of novel biomarkers to improve prognosis and guide therapy. ABCA1, an ATP‐binding cassette transporter involved in cholesterol efflux, has been implicated in tumorigenesis, yet its pan‐cancer roles and clinical relevance are not fully understood.

ABSTRACT Ten‐Eleven Translocation 2 (TET2) is an Fe(II)‐ and α‐ketoglutarate‐dependent dioxygenase that initiates active DNA demethylation by oxidizing 5‐methylcytosine (5mC). Beyond this enzymatic role, TET2 links DNA methylation dynamics with chromatin regulation, cellular metabolism, immune‐cell identity, and treatment response in a context ...

Acute and Chronic Leukemia in Sub‐Saharan Africa: A Systematic Review
ABSTRACT Background The four primary subgroups of leukemia include chronic myeloid leukemia (CML), acute myeloid leukemia (AML), acute lymphoblastic leukemia (ALL), and chronic lymphocytic leukemia (CLL). Very little is known about the regional variation of leukemia and subtypes in sub‐Saharan Africa (SSA).

TCF/LEF family transcription factors: Molecular landscape and prognostic scoring in pan‐cancer
This study provides a comprehensive molecular landscape of TCF/LEF family transcription factors across 33 cancer types. We developed a TCF/LEF‐based prognostic score that correlates with immune infiltration, metabolic reprogramming and drug sensitivity, offering a robust transcriptomic signature for predicting patient survival and therapeutic response.

Yy Abstract Background Cancer‐associated fibroblasts (CAFs) are key components of the breast cancer tumor microenvironment and mediate communication with tumor cells through extracellular vesicles carrying microRNAs (miRNAs). These miRNAs may influence breast cancer progression by regulating oncogenic and tumor‐suppressive pathways.

Interferon‐induced transmembrane (IFITM) function in cancer is set by four contextual coordinates, not by family membership. Acute interferon input drives major histocompatibility complex class I (MHC‐I)‐linked immunogenicity; chronic input drives programmed death‐ligand 1 (PD‐L1)‐coupled resistance.

Passenger variants as pan‐clonal markers of pediatric AML: Implications for molecular MRD
Abstract A streamlined, sensitive, and universal molecular method for MRD detection in pediatric AML remains an unmet clinical need. Here, we propose a novel approach based on tracking somatic non‐coding passenger variants. Using whole‐genome sequencing (WGS) of diagnostic bone marrow DNA, we identify patient‐specific somatic passenger variants, which ...

Clonal and genomic determinants of e1a2 BCR::ABL1 chronic myeloid leukemia: A Fi‐LMC study
Abstract The rarer p190 (e1a2) transcript in chronic myeloid leukemia (CML) is associated with atypical presentations; yet, its biological basis remains poorly understood. Using a cohort of 60 patients including 42 chronic phase patients age‐matched 1:1 with 42 e13a2/e14a2 patients in the chronic phase, we investigated the clinical, genomic, and clonal

ABSTRACT Acute myeloid leukemia (AML) is an aggressive hematologic malignancy associated with a high risk of relapse. Leukemia cutis is characterized by the presence of neoplastic leukocytes infiltrating the skin. Here, we present a previously treated AML patient who relapsed with isolated leukemia cutis without bone marrow involvement. Fluorescence in
