Pharmacokinetics and pharmacodynamics of intravenous and oral (S)‐ketamine: Investigating metabolite contribution to subjective effects

open access: yesBritish Journal of Clinical Pharmacology, Volume 92, Issue 7, Page 2260-2271, July 2026.
Aims Oral administration of (S)‐ketamine for treatment‐resistant depression (TRD), as alternative to the registered intranasal or off‐label intravenous administrations, has high potential. However, it is characterized by an extensive first‐pass metabolism, resulting in low (S)‐ketamine exposure and high levels of active metabolites, including (S ...

Nonlinear Pharmacokinetics of 5-Methoxy-N,N-dimethyltryptamine in Mice [PDF]

open access: yesDrug Metabolism and Disposition, 2011
5-Methoxy-N,N,-dimethyltryptamine (5-MeO-DMT), an abused serotonergic indolealkylamine drug, was placed into Schedule I controlled substance status in the United States as of January 19, 2011. In previous studies, we have shown the impact of monoamine oxidase A and cytochrome P450 2D6 enzymes on 5-MeO-DMT metabolism and pharmacokinetics.

Exploring Endogenous Tryptamines: Overlooked Agents Against Fibrosis in Chronic Disease? A Narrative Review

open access: yesLivers
Long regarded as illicit substances with no clinical value, N-dimethylated tryptamines—such as N,N-dimethyltryptamine, 5-methoxy-N,N-dimethyltryptamine, and bufotenine—have been found to produce naturally in a wide variety of species, including humans ...

Preclinical models for evaluating psychedelics in the treatment of major depressive disorder

open access: yesBritish Journal of Pharmacology, Volume 183, Issue 14, Page 3970-3991, July 2026.
Psychedelic drugs have seen a resurgence in interest as a next generation of psychiatric medicines with potential as rapid‐acting antidepressants (RAADs). Despite promising early clinical trials, the mechanisms which underlie the effects of psychedelics are poorly understood.

The Australia story: Current status and future challenges for the clinical applications of psychedelics

open access: yesBritish Journal of Pharmacology, Volume 183, Issue 14, Page 4048-4057, July 2026.
Abstract The past decade has seen a huge increase in clinical research with psychedelic drugs and 3,4‐methylenedioxymethamphetamine (MDMA), which have revealed great potential for treating mental health conditions.
