The dolichol pathway of N-linked glycosylation

Biochimica Et Biophysica Acta - General Subjects, 1999
The oligosaccharide substrate for the N-linked protein glycosylation is assembled at the membrane of the endoplasmic reticulum. Dolichyl pyrophosphate serves as a carrier in this biosynthetic pathway. In this review, we discuss the function of the lipid carrier dolichol in oligosaccharide assembly and give an overview of the biosynthesis of the ...

The Effects of N-Linked Glycosylation on SLC6 Transporters

Journal of Chemical Information and Modeling, 2022
Abstract Membrane transporters of the solute carrier 6 (SLC6) family mediate various physiological processes by facilitating the translocation of amino acids, neurotransmitters, and other metabolites. In the human body, these transporters are tightly controlled through various post-translational modifications with ...

A mathematical model of N‐linked glycosylation

Biotechnology and Bioengineering, 2005
AbstractMetabolic engineering of N‐linked oligosaccharide biosynthesis to produce novel glycoforms or glycoform distributions of a recombinant glycoprotein can potentially lead to an improved therapeutic performance of the glycoprotein product.

N-Linked Glycosylation of Mouse Adiponectin

Hormone and Metabolic Research, 2011
Adiponectin is an insulin-sensitizing adipokine with antidiabetic, anti-atherogenic, anti-inflammatory, and cardioprotective properties. Previously, some types of posttranslational modification on adiponectin have been reported. In this study, we demonstrate that mouse adiponectin protein migrated as 2 bands on SDS-PAGE gel.

Balancing N-linked glycosylation to avoid disease

Biochimie, 2001
Complete loss of N-glycosylation is lethal in both yeast and mammals. Substantial deficiencies in some rate-limiting biosynthetic steps cause human congenital disorders of glycosylation (CDG). Patients have a range of clinical problems including variable degrees of mental retardation, liver dysfunction, and intestinal disorders.

Inhibition of N‐Linked Glycosylation

Current Protocols in Protein Science, 1994
AbstractThis unit describes the use of inhibitors in cultured cells to prevent N‐linked glycosylation of proteins to yield glycoproteins containing missing or altered chains. This approach is useful for examining potential functional role(s) of oligosaccharides on specific proteins or intact cells.

N-Linked glycosylation

2011
This chapter discusses N-linked glycosylation. N-linked glycans, and the biosynthetic pathways that generate them, act as the cornerstone of our understanding of mammalian glycobiology. In spite of their diversity, all N-linked glycans on glycoproteins are generated through a common pathway, which starts in the endoplasmic reticulum with the transfer ...
