Transient Sleep Deprivation Induces Persistent Auditory Neuropathy via ROS‐Initiated Neuroinflammation and BK Channel Suppression

open access: yesAdvanced Science, EarlyView.
Sleep disturbance severity closely tracks hearing loss in a clinical cohort, yet the mechanistic link remains unclear. Acute sleep deprivation is shown to trigger transient cochlear oxidative stress that switches into a self‐sustaining neuroinflammatory state, suppressing BK channels and causing irreversible synaptopathy.

Arabidopsis HEAT SHOCK TRANSCRIPTION FACTORA1b overexpression enhances water productivity, resistance to drought, and infection [PDF]

open access: yes, 2013
Heat-stressed crops suffer dehydration, depressed growth, and a consequent decline in water productivity, which is the yield of harvestable product as a function of lifetime water consumption and is a trait associated with plant growth and development ...

The GPX3‐VCAM1 Axis Gates Pro‐Fibrotic Tubule Cell Fate in Hyperuricemic Nephropathy

open access: yesAdvanced Science, EarlyView.
Hyperuricemia induces NLRP3 inflammasome signaling activation and downregulation of GPX3 in proximal tubular epithelial cells, leading to oxidative stress–driven VCAM1 upregulation, emergence of pro‐fibrotic proximal tubule cells, macrophage recruitment, and renal fibrosis.

A DNA G‐Quadruplex‐Targeting Ligand Suppresses c‐MYC Expression: Implications for Gemcitabine‑Resistant Pancreatic Cancer

open access: yesAngewandte Chemie, EarlyView.
PQB‐4, a fluorescent small‐molecule ligand that targets c‐MYC DNA G‐quadruplexes (G4s), showed marked synergy with gemcitabine in resistant PANC‐1 cells (CI = 0.027). In vivo, the ligand reduced tumor weight by 44% in a PANC‐1 xenograft model. These results highlight the therapeutic potential of c‐MYC DNA G4‐targeting ligands for treating gemcitabine ...

Magnetically Guided Macrophage Immunobots Coordinate Iron‐Metabolic Cascades and Immunogenic Ferroptosis for Tumor Immunotherapy

open access: yesAdvanced Science, EarlyView.
Magnetically guided macrophage immunobots integrate active tumor targeting with immunometabolic therapy. After systemic administration, magnetic actuation enhances immunobot retention and tumor penetration, while LPS‐functionalized FePt microrollers sustain M1‐like polarization through inflammatory and iron‐amplified oxidative signaling and trigger ...

Recent advances in the structural molecular biology of Ets transcription factors: interactions, interfaces and inhibition. [PDF]

open access: yes, 2014
The Ets family of eukaryotic transcription factors is based around the conserved Ets DNA-binding domain. Although their DNA-binding selectivity is biochemically and structurally well characterized, structures of homodimeric and ternary complexes point to

Hederagenin Attenuates Morphine Addiction via GABAA1‐Mediated Modulation of Ca2+–CaMK–CREB Signaling in Ventral Tegmental Dopaminergic Neurons

open access: yesAdvanced Science, EarlyView.
Hederagenin is identified as a GABAA1‐targeting natural compound that suppresses morphine‐associated reward by restraining Ca2+–CaMK–cAMP–CREB signaling in ventral tegmental dopaminergic neurons. RVG29‐modified liposomal delivery further enhances brain accumulation and behavioral efficacy, providing a mechanistically defined strategy for natural ...
