A folate receptor‐targeted nanoplatform co‐targets lipid droplets and lysosomes to orchestrate ferroptosis, ICD, and STING activation for photo‐immunotherapy. Under light, it induces ferroptosis and ICD, releasing mtDNA that synergizes with a co‐delivered agonist to hyperactivate the STING pathway and remodel the tumor microenvironment.

This work proposes a novel strategy to fabricate unimolecular pyroptosis photoinitiators activated by deep‐red light at low light doses. It relies on a photosensitization mechanism to trigger nitric oxide (NO) release from benzisothiadiazole‐based N‐nitrosanilines without modification of nitro moieties, thereby boosting pyroptosis‐mediated ...

Antibody–Drug Conjugates and Peptide–Drug Conjugates: Current Understandings and Future Perspectives
The graphic summarizes the design principles, intracellular trafficking, clinical progress, challenges, and future directions of antibody–drug conjugates (ADCs) and peptide–drug conjugates (PDCs). ADCs consist of a tumor‐targeting antibody, a chemical linker, and a potent payload, whereas PDCs use peptide ligands to deliver cytotoxic, radionuclide, or ...

Near-Infrared Photoimmunotherapy: Photoactivatable Antibody–Drug Conjugates (ADCs)
Cancer treatment has been founded traditionally on the three approaches of surgery, radiation, and chemotherapy with the latter recognized as the obvious systemic treatment approach applicable to disease that has spread. Although significant progress has been made over nearly 100 years of developing systemic treatments, it remains clear that use of the
