Abstract Rat liver NAD kinase (ATP:NAD 2'-phosphotransferase, EC 2.7.1.23) was purified about 70-fold. The Michaelis-Menten constants (Km) for NAD and ATP were 8 x 10-4 m and 2 x 10-3 m, respectively. NAD kinase activity was markedly inhibited by NADH and also NADPH.

A Fluorescent Analog of Nicotinamide Adenine Dinucleotide [PDF]
Nicotinamide 1, N 6 -ethenoadenine dinucleotide, a fluorescent analog of the coenzyme nicotinamide adenine dinucleotide, has been synthesized by the reaction of chloroacetaldehyde with the coenzyme. The technical fluorescence emission maximum of the analog is 410 nm, upon excitation at 300 nm.

Circulating blood eNAMPT drives the circadian rhythms in locomotor activity and energy expenditure
Nicotinamide phosphoribosyl transferase (NAMPT) is a critical regulator of cellular nicotinamide adenine dinucleotide (NAD)+ levels. Here, the authors show that blood NAMPT contributes to the circadian rhythm generation of locomotor activity and energy ...

The Role of Pyruvate Dehydrogenase Kinase in Diabetes and Obesity [PDF]
The pyruvate dehydrogenase complex (PDC) is an emerging target for the treatment of metabolic syndrome. To maintain a steady-state concentration of adenosine triphosphate during the feed-fast cycle, cells require efficient utilization of fatty acid and ...

Improved catalytic activity of ruthenium–arene complexes in the reduction of NAD+ [PDF]
A series of neutral Ru-II half-sandwich complexes of the type [(eta(6)-arene)Ru(N,N')Cl] where the arene is para-cymene (p-cym), hexamethylbenzene (hmb), biphenyl (bip), or benzene (bn) and N,N' is N-(2-aminoethyl) -4-(trifluoromethyl)benzenesulfonamide (

It takes two to tango: NAD+ and sirtuins in aging/longevity control [PDF]
The coupling of nicotinamide adenine dinucleotide (NAD+) breakdown and protein deacylation is a unique feature of the family of proteins called ‘sirtuins.’ This intimate connection between NAD+ and sirtuins has an ancient origin and provides a ...

Nicotinamide Adenine Dinucleotide Metabolism and Neurodegeneration [PDF]
Nicotinamide adenine dinucleotide (NAD+) participates in redox reactions and NAD+-dependent signaling processes, which involve the cleavage of NAD+ coupled to posttranslational modifications of proteins or the production of second messengers. Either as a primary cause or as a secondary component of the pathogenic process, mitochondrial dysfunction and ...
