Nsp9 is a conserved accessory component of the coronaviral replication and transcription complex. It is the predominant substrate of nsp12’s nucleotidylation activity while also serving to recruit proteins required for viral 5’-capping.

The SARS-CoV-2 replication and transcription complex (RTC) provides RNA-dependent RNA polymerase (RdRp) activity. In its minimal composition, it consists of ‘non-structural proteins’, including nsp12 in complex with its protein cofactors: one copy of nsp7 and two of nsp8.
