Mutations in chikungunya virus nsP4 decrease viral fitness and sensitivity to the broad-spectrum antiviral 4'-Fluorouridine.

open access: yesPLoS Pathogens
Chikungunya virus (CHIKV) is an arthritogenic alphavirus that has re-emerged to cause large outbreaks of human infections worldwide. There are currently no approved antivirals for treatment of CHIKV infection.

Recent Increase in Rotavirus Gastroenteritis After Five Years of Low Activity in Hokkaido Prefecture, Japan, Associated With Equine‐Like G3P[8] and G8P[8] Strains, 2025

open access: yesJournal of Medical Virology, Volume 98, Issue 5, May 2026.
ABSTRACT Rotavirus gastroenteritis decreased substantially worldwide during the COVID‐19 pandemic, and activity remained minimal in Hokkaido Prefecture, Japan, for 5 years after 2020. Hospital‐based surveillance in Hokkaido since July 2019 identified the first resurgence of rotavirus gastroenteritis in 2025.

Predicted binding interface between coronavirus nsp3 and nsp4 [PDF]

open access: yes, 2022
Abstract Double membrane vesicles (DMVs) in coronavirus-infected cells feature pores that span both membranes. DMV pores were observed to have six-fold symmetry and include the nsp3 protein. Co-expression of SARS-CoV nsp3 and nsp4 induces DMV formation, and elements of nsp3 and nsp4 have been identified that are essential for membrane ...

Minimal Polymerase-Containing Precursor Required for Chikungunya Virus RNA Synthesis

open access: yesViruses
Alphaviruses pose a growing global health threat, with Chikungunya virus (CHIKV) epidemics ongoing. Although several CHIKV vaccine candidates have progressed to late-stage clinical evaluation, none have yet achieved licensure or widespread availability ...

The Molecular Chaperone Calnexin Interacts with the NSP4 Enterotoxin of Rotavirus In Vivo and In Vitro [PDF]

open access: yesJournal of Virology, 1998
ABSTRACT Calnexin is an endoplasmic reticulum (ER)-associated molecular chaperone proposed to promote folding and assembly of glycoproteins that traverse the secretory pathway in eukaryotic cells. In this study we examined if calnexin interacts with the ER-associated luminal (VP7) and transmembrane (NSP4) proteins of rotavirus.

Chloride secretion induced by rotavirus is oxidative stress-dependent and inhibited by Saccharomyces boulardii in human enterocytes. [PDF]

open access: yesPLoS ONE, 2014
Rotavirus (RV) infection causes watery diarrhea via multiple mechanisms, primarily chloride secretion in intestinal epithelial cell. The chloride secretion largely depends on non-structural protein 4 (NSP4) enterotoxic activity in human enterocytes ...

Alphaviral Capsid Proteins Inhibit Stress Granule Assembly via Competitive RNA Binding With G3BP1

open access: yesAdvanced Science, Volume 13, Issue 20, 9 April 2026.
Stress granules exert antiviral functions. This study illustrates a conserved function of alphaviral capsid proteins in modulating stress granules. Oligomerization mediated by a helical motif coupled with a positively charged intrinsically disordered region (IDR) directly competes with G3BP1 for RNA binding, thereby disrupting G3BP1‐RNA liquid–liquid ...

Coronavirus Nsp3 Hijacks CLTC to Modulate Autophagosome Nucleation for Promoting DMV Formation and Viral Replication

open access: yesAdvanced Science, Volume 13, Issue 24, 27 April 2026.
In wild‐type cells, FIPV infection recruits CLTC to nsp3, facilitates DMV biogenesis and block autophagic flux to promote viral replication. CLTC knockout impairs autophagosome nucleation by reducing Beclin1–ATG14 complex expression. This disrupts the formation of autophagic precursor membranes, thereby preventing their hijacking by nsp3 for DMV ...

Increased immunogenicity and protective efficacy of influenza M2e fused to a tetramerizing protein.

open access: yesPLoS ONE, 2012
The ectodomain of the matrix 2 protein (M2e) of influenza A virus represents an attractive target for developing a universal influenza A vaccine, with its sequence being highly conserved amongst human variants of this virus.
