Murine hepatitis virus nsp4 N258T mutants are not temperature-sensitive

Coronavirus replicase nsp4 is critical for virus-induced membrane modifications. An nsp4 mutant (N258T) of murine hepatitis virus (MHV) has been reported to be temperature-sensitive (ts) and to alter membrane targeting. We engineered and recovered all four possible codon variants of N258T in the cloned MHV-A59 background.

Abstract The SARS‐CoV‐2 main protease (Mpro) is essential for viral replication and functions as a homodimer, with dimerization being critical for catalytic activity. Mpro contains an unusually high number of cysteine residues. Among these, C117 and the catalytic nucleophile C145 can form a reversible disulfide bond under oxidative conditions.

The rotavirus nonstructural glycoprotein NSP4 possesses membrane destabilization activity [PDF]

During a unique morphogenetic process, rotaviruses obtain a transient membrane envelope when newly synthesized subviral particles bud into the endoplasmic reticulum (ER). As rotavirus particles mature, they lose their transient membrane and a layer of the glycoprotein VP7 forms the virion outer capsid shell.

ABSTRACT Group A rotavirus (RVA) genotype G3P[8] strains typically carry a Wa‐like genomic backbone, but DS‐1‐like G3P[8] reassortants have disseminated globally since 2013. In Sabah, Malaysia, surveillance in 2018–2019 identified frequent G3P[8] strains with a short RNA electropherotype, consistent with DS‐1‐like backbones.

Diarrhea Induction by Rotavirus NSP4 in the Homologous Mouse Model System

Comparison of the NSP4 amino acid sequences from 31 strains of mammalian rotaviruses revealed the presence of four distinct NSP4 alleles; i.e., the Wa, KUN, AU-1, and EW alleles. The EW allele consists only of NSP4s from murine rotavirus strains and is divergent from other NSP4 alleles from the evolutionary perspective.
