Analysis and purification of peptide nucleic acids by denaturing PAGE
ELECTROPHORESIS, 2007AbstractA flexible and convenient protocol for the analysis and purification of peptide nucleic acid (PNA) oligomers and PNA‐peptide chimeras by denaturing PAGE is described. Vertical slab gel electrophoresis, 26% in polyacrylamide and 8 M urea at pH 3, was suitable for analysis of oligomers ranging in size from tetramers (4‐mers) to tetradodecamers ...

Numerical analysis of thermal denaturation of nucleic acids
Analytical Biochemistry, 1978Denaturation of DNA molecules by stepwise incrementation of the temperature leads to melting profiles showing a fine structure, composed of individual melting modes. A method is described by which quantitative physical information brought by the modes can be extracted from the melting profile.

Denaturation bubble-mediated two-stage isothermal nucleic acid amplification in a single closed tube
Chemical Communications, 2023A two-stage isothermal amplification method termed rolling cycle strand exchange amplification (RC-SEA) was established to accomplish ssRNA and dsDNA analysis using only one DNA polymerase in a single closed tube at a constant temperature.

Pyridine: a Denaturant or Stabilizer of Spherical Nucleic Acids?
Analytical Chemistry, 2017The spotlighted dual functions of pyridine as a denaturant and as a stabilizer for duplex DNA are thoroughly investigated using spherical nucleic acids (SNAs). At neutral pH, pyridine destabilizes the duplex interconnects of assembled SNAs, resulting in a gradual decrease in their melting temperature (Tm) as a function of the pyridine concentration ...

Abstract Sedimentation studies indicate that native DNA and DNA denatured by dilution with water below the critical concentration zone have similar sedimentation coefficients and sedimentation coefficient concentration dependence. This is similar to the findings for alkali and thermally denatured DNA.

Small-volume cuvette for high-resolution thermal denaturation of nucleic acids
Analytical Biochemistry, 1979Abstract A small-volume cuvette and hoider makes it possible to obtain useful high-resolution thermal denaturation profiles on as little as submicrogram quantities of DNA and routine results with approximately 2 μg of DNA. The reduced sample size is especially important in the thermal characterization of DNA restriction fragments.

Theoretical Aspects of Thermal Denaturation of Nucleic Acids
1998Information on mutations (as measured by D/TGGE) can be significantly improved by the use of a theoretical description of the thermal denaturation of nucleic acids.

Microfluidic chip of fast DNA hybridization using denaturing and motion of nucleic acids
ELECTROPHORESIS, 2008AbstractThis study presents the effect of fluidic temperatures and velocities on improving DNA hybridization. The efficiency of hybridization could be improved by introducing elevated temperature in the hot region and velocity in the cold region. Compared with the conventional methods, this hybridization microchip was able to increase the hybridization

Effects of cupric ions on thermal denaturation of nucleic acids
Journal of Molecular Biology, 1965The midpoint of thermal denaturation of sodium DNA and RNA decreases with increasing concentration of cupric ions. With Micrococcus lysodeikticus DNA, the Tm† is 69°C without cupric ions and 36°C with 0·1 m M -cupric ions in 5 m M -sodium chloride. In a solvent consisting of 5 m M -sodium chloride and 0·1 m M -cupric ions, Tm values of several DNA's ...

Fast DNA hybridization chip using denature and motion of nucleic acids
2010 5th International Microsystems Packaging Assembly and Circuits Technology Conference, 2010We demonstrate that the efficiency of DNA hybridization could be improved by introducing elevated temperature in the hot region and higher velocities in the cold region of a microfluidic chip. Compared with the conventional methods, this hybridization microchip was shown to increase the hybridization signal 4.6-fold within 30 minutes using a 1.4 kb ...
