Aldehyde or Hydrate? Investigation into the Oxidation of 5‐Formylcytosine Derivatives Using a Computational and Experimental Approach

open access: yesChemBioChem, EarlyView.
Hydrate‐mediated oxidation of 5‐formyl nucleobases by the iron(IV)‐oxido complex proceeds via geminal diol intermediates, accelerating hydrogen‐atom transfer and revealing aza‐dependent kinetic and mechanistic pathways. This study investigates the oxidation of 5‐hydroxymethyl and 5‐formyl nucleobases using an iron(IV)‐oxido complex that mimics the ...

Model‐Based Optimization of Fed‐Batch In Vitro Transcription

open access: yesChemBioChem, EarlyView.
Using a mathematical model for the enzyme kinetics and solution thermodynamics of the in vitro transcription reaction, new optimized strategies are developed for fed‐batch transcription that can decrease the resource costs of mRNA synthesis while maintaining critical quality attributes for efficacy in vivo.

Ratiometric Detection of pH‐Induced i‐Motif Folding Based on a Dual Emissive Cytosine Analog

open access: yesChemBioChem, EarlyView.
A dual‐emissive cytosine analog enables ratiometric fluorescence detection of i‐motif DNA folding. This study reveals a sensitive method to monitor pH‐dependent i‐motif conformational changes, providing a valuable tool for investigating DNA secondary structures in vitro.

Biocompatible Chemistry: A Plug‐and‐Play Toolbox for Chemical Biology Research

open access: yesChemBioChem, EarlyView.
The “plug‐and‐play” nature of biocompatible chemistry makes it a powerful toolbox for investigating complicated biological systems and producing drug leads for clinical usages. This review summarizes the commonly used biocompatible reactions and the recent advances of their representative applications in chemical biology research, thus highlighting the
