Effects of obidoxime chloride on native and sarin-poisoned frog neuromuscular junctions

Pflgers Archiv European Journal of Physiology, 1984
The effect of the oxime reactivator obidoxime chloride (obidoxime) on single frog neuromuscular junctions has been studied in order to clarify its action on the acetylcholine receptor (AChR) and on the acetylcholine esterase (AChE), both before and after blocking its enzymatic activity with the organophosphorus compound sarin. Experiments iontophoretic

Efficacy of Pralidoxime Iodide and Obidoxime Dichloride as Antidotes in Diazinon-Poisoned Goslings

Avian Diseases, 1976
The antidotal efficacy of pralidoxime iodide and obidoxime dichloride was investigated in goslings poisoned by a supralethal dose of the organophosphorus insecticide diazinon. Various doses of both drugs were administered by intramuscular injection when the poisoned birds were unable to walk. Pralidoxime at 100 mg/kg brought about a complete and speedy

Spectrophotometric determination of obidoxime chloride as its Pd(II) complex in water and injections

Journal of Pharmaceutical and Biomedical Analysis, 1987
It has been established that obidoxime chloride reacts with palladium(II) chloride in the pH range 3.9-8.0 and forms a yellow water-soluble (1:1) complex with maximum absorbance at 355 nm. By applying the methods of Sommer, Nash and Job involving non-equimolar solutions the conditional stability constant of the complex, at the optimum pH of 6.3 and an ...

Reactivation of nerve agent-inhibited human acetylcholinesterase by obidoxime, HI-6 and obidoxime+HI-6: Kinetic in vitro study with simulated nerve agent toxicokinetics and oxime pharmacokinetics

Toxicology, 2016
Despite extensive research for decades no effective broad-spectrum oxime for the treatment of poisoning by a broad range of nerve agents is available. Previous in vitro and in vivo data indicate that the combination of in service oximes could be beneficial.

Beneficial late administration of obidoxime in malathion poisoning.

Veterinary and human toxicology, 2003
Early treatment of organophosphate (OP) poisoning with oximes results in reactivation of acetylcholinesterase and patient recovery. Data on efficacy of late administration of oximes, particularly obidoxime, is limited. A 42-y old woman swallowed 60 ml of 50% malathion in a suicide attempt. Characteristic muscarinic, nicotinic and central manifestations

Cholinesterase status, pharmacokinetics and laboratory findings during obidoxime therapy in organophosphate poisoned patients

Human & Experimental Toxicology, 1997
1 The effectiveness of oxime therapy in organophos phate poisoning is still a matter of debate. It appears, however, that the often cited ineffectiveness of oximes may be due to inappropriate dosing. By virtue of in vitro findings and theoretical considerations we concluded in the preceding paper that oximes should preferably be administered by ...

Obidoxime Augments the Positive Inotropic Effect of Phosphamidon on the Isolated Working Rat Heart

Pharmacology & Toxicology, 1992
Abstract: The present study was designed to determine modulation of the direct inotropic effect of an anticholinesterase organophosphorus compound, phosphamidon, by the reactivator obidoxime. We investigated the effects of phosphamidon (n = 9), obidoxime (n = 5), and their combination (n = 5) on the mechanical and energetic indices of left ventricular

Efficacy of atropine sulfate/obidoxime chloride co-formulation against sarin exposure in guinea pigs

Chemico-Biological Interactions, 2018
The efficacy and pharmacokinetics of the aqueous co-formulation contents of the Trobigard™ (atropine sulfate, obidoxime chloride) auto-injector were evaluated in a sarin exposed guinea pig model. Two subcutaneous (sc) sarin challenge doses were evaluated in guinea pigs instrumented with brain and heart electrodes for electroencephalogram (EEG) and ...

Pharmacokinetics of [14C]-Sarin and Its Changes by Obidoxime and Pralidoxime

1986
Sarin was invented by Schrader in 1937 (6). It was one of the first examples of an organophosphate; it was later stockpiled with other similar compounds (soman, tabun and VX) as nerve gases for warfare. Of all the other 2000 organophosphorus compounds synthesized as insecticides, only several have been used to a major extent in agriculture.
