The tissue‐specific transcription factors HNF1α and HNF1β are closely related homeodomain proteins conserved in vertebrate evolution. Heterozygous mutations in human HNF1β but not in HNF1α genes are associated with kidney malformations. Overexpression of HNF1β in Xenopus embryos leads to defective pronephros development, while HNF1α has no effect.

2006The transcription factor Pax8 is involved in the expression of the thyroid-specific genes that are considered markers of the differentiated phenotype. The regulation of Pax8 activity is likely to play a key role in thyroid differentiation as well as in thyroid malignant transformation.

Mutations of Wilms’ tumour gene 1 (WT1) in AML are rare and WT1 expression may be increased by other transcription factors. PAX2, PAX8 and GATA‐1 are known physiological regulators of WT1.
