Plasmodium falciparum gametogenesis essential protein 1 (GEP1) is a transmission‐blocking target

open access: yesFEBS Letters, EarlyView.
This study shows Plasmodium falciparum GEP1 is vital for activating sexual stages of malarial parasites even independently of a mosquito factor. Knockout parasites completely fail gamete formation even when a phosphodiesterase inhibitor is added. Two single‐nucleotide polymorphisms (V241L and S263P) are found in 12%–20% of field samples.

CHREST tutorial: Simulations of human learning [PDF]

open access: yes, 2004
CHREST (Chunk Hierarchy and REtrieval STructures) is a comprehensive, computational model of human learning and perception. It has been used to successfully simulate data in a variety of domains, including: the acquisition of syntactic categories, expert

Chimeric 16S rRNA sequence formation and detection in Sanger and 454-pyrosequenced PCR amplicons.

open access: yesGenome Research, 2011
Bacterial diversity among environmental samples is commonly assessed with PCR-amplified 16S rRNA gene (16S) sequences. Perceived diversity, however, can be influenced by sample preparation, primer selection, and formation of chimeric 16S amplification ...

Real-time RT-PCR in COVID-19 detection: issues affecting the results

open access: yesExpert Review of Molecular Diagnostics, 2020
Due to the rapid spread and increasing number of coronavirus disease 19 (COVID-19) cases caused by a new coronavirus, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), rapid and accurat...

4‐nitrobenzoate inhibits 4‐hydroxybenzoate polyprenyltransferase in malaria parasites and enhances atovaquone efficacy

open access: yesFEBS Letters, EarlyView.
Atovaquone is an antimalarial requiring potentiation for sufficient efficacy. We pursued strategies to enhance its activity, showing that 4‐nitrobenzoate inhibits 4‐hydroxybenzoate polyprenyltransferase, decreasing ubiquinone biosynthesis. Since atovaquone competes with ubiquinol in mitochondria, 4‐nitrobenzoate facilitates its action, potentiating ...

EPAM/CHREST tutorial: Fifty years of simulating human learning [PDF]

open access: yes, 2009
Generating quantitative predictions for complex cognitive phenomena requires precise implementations of the underlying cognitive theory. This tutorial focuses on the EPAM/CHREST tradition, which has been providing significant models of human ...

Stability issues of RT‐PCR testing of SARS‐CoV‐2 for hospitalized patients clinically diagnosed with COVID‐19

open access: yesJournal of Medical Virology, 2020
In this study, we collected a total of 610 hospitalized patients from Wuhan between February 2, 2020, and February 17, 2020. We reported a potentially high false negative rate of real‐time reverse‐transcriptase polymerase chain reaction (RT‐PCR) testing ...

Linked dimers of the AAA+ ATPase Msp1 reveal energetic demands and mechanistic plasticity for substrate extraction from lipid bilayers

open access: yesFEBS Letters, EarlyView.
Cells must clear mislocalized or faulty proteins from membranes to survive. The AAA+ ATPase Msp1 performs this task, but dissecting how its six subunits work together is challenging. We engineered linked dimers with varied numbers of functional subunits to reveal how Msp1 subunits cooperate and use energy to extract proteins from the lipid bilayer ...

An investigation into the effect of ageing on expert memory with CHREST [PDF]

open access: yes, 2007
CHREST is a cognitive architecture that models human perception, learning, memory, and problem solving, and which has successfully simulated numerous human experimental data on chess. In this paper, we describe an investigation into the effects of ageing
