The discovery and development of ensifentrine: A novel inhaled dual PDE3/4 inhibitor having ‘bifunctional’ bronchodilator and anti‐inflammatory activity

open access: yesBritish Journal of Pharmacology, EarlyView.
Abstract Ensifentrine (formerly called RPL 554) is a novel, first in class, inhaled bifunctional dual PDE3/4 inhibitor for the treatment of chronic respiratory diseases. Ensifentrine exhibits both bronchodilation via PDE3 inhibition and anti‐inflammatory activity via PDE4 inhibition.

Optimization of Substituted N-3-Benzylimidazoquinazolinone Sulfonamides As Potent and Selective PDE5 Inhibitors [PDF]

open access: yes, 2000
A previous report from these laboratories identified the N-3-benzylimidazoquinazolinone nucleus as a more selective PDE5 inhibitor template compared to the pyrazolopyrimidine of sildenafil.

Shared Decision‐Making During Pulmonary Arterial Hypertension Therapy Selection: A Patient and Clinician Survey

open access: yesPulmonary Circulation, Volume 16, Issue 3, September 2026.
ABSTRACT Selection of therapy for pulmonary arterial hypertension (PAH) requires tradeoffs among disease severity, therapeutic benefit, adverse effects, treatment burden, quality of life, and patient values. Although professional societies endorse shared decision‐making (SDM) in this context, empirical data describing SDM in PAH are limited.

Furoyl and Benzofuroyl Pyrroloquinolones as Potent and Selective PDE5 Inhibitors for Treatment of Erectile Dysfunction [PDF]

open access: yes, 2016
Synthesis of furoyl and benzofuroyl pyrroloquinolones as potent and selective PDE5 inhibitors was reported. Their in vitro potencies in inhibiting PDE5 and selectivity in inhibiting other PDE isozymes (PDE1−4 and PDE6) were evaluated.

Substituted Pyrazolopyridopyridazines as Orally Bioavailable Potent and Selective PDE5 Inhibitors:  Potential Agents for Treatment of Erectile Dysfunction [PDF]

open access: yes, 2016
Novel pyrazolopyridopyridazine derivatives have been prepared as potent and selective PDE5 inhibitors. Compound 6 has been identified as a more potent and selective PDE5 inhibitor than sildenafil (1). It is as efficacious as sildenafil in in vitro and in

The Clinical and Economic Burden of Pulmonary Arterial Hypertension in Israel, 2014–2022: A Retrospective Analysis of Healthcare Costs and Utilization

open access: yesPulmonary Circulation, Volume 16, Issue 3, September 2026.
ABSTRACT Pulmonary arterial hypertension (PAH) is a rare, progressive disease with high morbidity and mortality. Real‐world data from Israel are scarce. This study aimed to estimate the incidence, describe treatment patterns, and quantify healthcare utilization and costs associated with PAH in Israel.

Revisiting Controversial Areas in the Management of Premature Ejaculation

open access: yesTrends in Urology &Men's Health, Volume 17, Issue 4, August 2026.
ABSTRACT The cause of premature ejaculation (PE) is often multifactorial in nature, and effective management of PE is complex and should involve multimodal approaches. This narrative review examines the ongoing controversies surrounding key areas in the diagnosis and treatment of PE with an emphasis on future research directions to align clinical care ...
