Selective Phosphorylation of Phenols and Benzenediols by the Kinase PsiK and Variants Thereof

PsiK from Psilocybe cubensis phosphorylates a diverse set of substituted phenols and benzenediols beyond its native substrate. High‐throughput screening of active site mutant libraries using DESI‐MS enabled protein engineering of PsiK, further expanding its substrate scope.

Single‐Molecule Protein Profiling Using Nanopores and Dimeric Aptamer‐Modified DNA Carriers

We developed a nanopore‐based biosensor for single‐molecule protein profiling using aptamer‐modified DNA carrier dimerization strategy, enabling selective and ultrasensitive detection of proteins and protein complexes. This approach promises advances in early‐stage clinical diagnostics and precision medicine.

Decoy DNA Protects Molecular Tension Probes from DNase Degradation

DNA‐based probes are rapidly degraded by DNases in live‐cell environments, limiting their use. We introduce a decoy DNA strategy, where an excess of unmodified double‐stranded DNA acts as a competitive substrate for DNases, protecting functional DNA probes and significantly extending their stability for long‐term live‐cell studies.
