Endocytic Control of Cell‐Autonomous and Non‐Cell‐Autonomous Functions of p53

open access: yesAdvanced Science, EarlyView.
NUMB Ex3‐containing isoforms localize to the plasma membrane, where they recruit p53 through SNX9 and direct it to multivesicular bodies and exosomes. Exported p53 is taken up by neighboring cells and activates nuclear programs, revealing an intercellular, exosome‐based pathway that might help establish a tumor‐suppressive microenvironment.

Determination of VPS34/PIK3C3 Activity in vitro Utilising 32P-γATP

open access: yesBio-Protocol, 2016
VPS34 is the only class III phosphatidylinositol-3-kinase (PI3K) in mammalian cells and produces the vast majority of cellular phosphatidylinositol-3-phosphate [PI(3)P].

Inportance of Phosphatidylinositol 3-Phosphate in Sporulation ofSchizosaccharomyces pombe [PDF]

open access: yesBioscience, Biotechnology, and Biochemistry, 2003
In Schizosaccharomyces pombe, Pik3p phosphorylates phosphatidylinositol (PI) to produce PI 3-P, which is further phosphorylated by Ste12p to yield PI 3,5-P2. Pik3p is required for both conjugation and sporulation. To test which of PI 3-P and PI 3,5-P2 is required for sporulation, diploid cells defective in production of PI 3,5-P2 were used.

Delivery of Pleckstrin‐Homology Domains Suppresses PI3K/Akt Signaling and Breast Cancer Metastasis

open access: yesAdvanced Science, EarlyView.
Current therapies curb tumor growth but not metastasis. Obscurin, a giant metastasis suppressor lost in breast cancer, restrains PI3K/Akt signaling but is impractical to restore. We deploy a mini‐obscurin, comprising the obscurin‐PH‐domain, which sequesters PI3K‐p85, potently suppressing invasion and metastasis.

The casein kinases Yck1p and Yck2p act in the secretory pathway, in part, by regulating the Rab exchange factor Sec2p. [PDF]

open access: yes, 2016
Sec2p is a guanine nucleotide exchange factor that activates Sec4p, the final Rab GTPase of the yeast secretory pathway. Sec2p is recruited to secretory vesicles by the upstream Rab Ypt32p acting in concert with phosphatidylinositol-4-phosphate (PI(4)P).

Making autophagosomes: Localized synthesis of phosphatidylinositol 3-phosphate holds the clue [PDF]

open access: yesAutophagy, 2008
Autophagy presents a topological challenge for the cell because it requires delivery of cytosolic material to the lumen of a membrane-bound compartment, the lysosome. This is solved in an ingenious way by the formation of a double-membrane vesicle, the autophagosome, which captures cytosolic proteins and organelles during its transformation from a ...

Integration of Spatiotemporal Multi‐Omics in Peach Fruit Unravels a Metabolic Niche and the Genetic Basis of Trichome‐Mediated Stress Adaptation

open access: yesAdvanced Science, EarlyView.
This study constructed the first spatiotemporal multi‐omics map of peach fruit and discovered a key candidate gene that synergistically regulates trichome development and drought tolerance through the jasmonic acid signaling pathway, providing insights into the coupling mechanism between development and stress resistance.

An emerging role for PI5P in T cell biology

open access: yesFrontiers in Immunology, 2013
Phosphoinositides are critical regulators in cell biology. Phosphatidylinositol 4,5-bisphosphate, also known as PI(4,5)P2 or PIP2, was the first variety of phosphoinositide to enter in the T cell signaling scene.

Kytococcus aerolatus sp. nov., isolated from indoor air in a room colonized with moulds [PDF]

open access: yes, 2009
A Gram-positive, coccoid bacterial isolate (02-St-019/1(T)), forming beige pigmented colonies was obtained from an indoor air sample. Based on 16S rRNA gene sequence similarity studies it was determined that this isolate 02-St-019/1(T) belonged to the ...

Nanoscale Spatial Organization of ARC High‐ and Low‐Order Assemblies at Excitatory Synapses

open access: yesAdvanced Science, EarlyView.
ARC (Activity‐Regulated Cytoskeleton‐Associated protein) mediates synaptic plasticity by forming nanoscale assemblies in neurons. Using super‐resolution microscopy and time‐resolved anisotropy with targeted tagging, the study reveals low‐order ARC assemblies at synapses colocalizing with AMPARs, semi‐circular structures at endocytic zones, and 60–80 nm
