The existence of a PIP5K family of enzymes has been suggested by Western blotting and purification of numerous PIP5Ks from various tissues and cell types. The erythrocyte has at least two PIP5Ks, named PIP5KI and PIP5KII, while the brain appears to have even more isoforms.

AbstractType I phosphatidylinositol 4‐phosphate 5‐kinase (PI4P5K) catalyzes the phosphorylation of phosphatidylinositol 4 phosphate [PI(4)P] at carbon 5, producing phosphatidylinositol 4,5 bisphosphate [PI(4,5)P2]. Phosphatidic acid (PA) activates PI4P5K in vitro and plays a central role in the activation of PIP5K pathways in vivo.

Inositol lipid kinases are perhaps the easiest and most straightforward enzymes in the phosphoinositide pathway to analyze. In addition to monitoring lipid kinase-specific activity, lipid kinase assays can be used to quantify the inositol lipids present in isolated membranes (Jones et al., Methods Mol Biol 462:75-88, 2009).

Phosphatidylinositol 4,5-bisphosphate (PIP2) is enzymatically produced when high speed supernatant fraction from bovine retina is incubated with [gamma-32P]ATP and phosphatidylinositol 4-phosphate (PIP) as substrates. Exogenously added PIP2 inhibits PIP kinase activity 50% at equimolar concentrations of product and substrate.
