PIP4Ks Suppress Insulin Signaling through a Catalytic-Independent Mechanism

open access: yesCell Reports, 2019
Summary: Insulin stimulates the conversion of phosphatidylinositol-4,5-bisphosphate (PI(4,5)P2) to phosphatidylinositol-3,4,5-trisphosphate (PI(3,4,5)P3), which mediates downstream cellular responses.

A conserved function in phosphatidylinositol metabolism for mammalian Vps13 family proteins. [PDF]

open access: yesPLoS ONE, 2015
The Vps13 protein family is highly conserved in eukaryotic cells. In humans, mutations in the gene encoding the family member VPS13A lead to the neurodegenerative disorder chorea-acanthocytosis.

Osh6 Revisited: Control of PS Transport by the Concerted Actions of PI4P and Sac1 Phosphatase

open access: yesFrontiers in Molecular Biosciences, 2021
Osh6, a member of the oxysterol-binding protein–related protein (ORP) family, is a lipid transport protein that is involved in the transport of phosphatidylserine (PS) between the endoplasmic reticulum (ER) and the plasma membrane (PM).

Purification and characterization of phosphatidylinositol 4-phosphate 5-kinases [PDF]

open access: yesBiochemical Journal, 1992
We detail the purification and characterization of three distinct isoforms of PtdIns4P 5-kinase present in bovine brain. One of these, PtdIns4P 5-kinase C, was purified to apparent homogeneity, and SDS/PAGE analysis demonstrated a single polypeptide and molecular mass 53 KDa.

Membrane-mediated dimerization potentiates PIP5K lipid kinase activity

open access: yeseLife, 2022
The phosphatidylinositol 4-phosphate 5-kinase (PIP5K) family of lipid-modifying enzymes generate the majority of phosphatidylinositol 4,5-bisphosphate [PI(4,5)P2] lipids found at the plasma membrane in eukaryotic cells.

Characterization of VPS34-IN1, a selective inhibitor of Vps34, reveals that the phosphatidylinositol 3-phosphate-binding SGK3 protein kinase is a downstream target of class III phosphoinositide 3-kinase [PDF]

open access: yes, 2014
The Vps34 (vacuolar protein sorting 34) class III PI3K (phosphoinositide 3-kinase) phosphorylates PtdIns (phosphatidylinositol) at endosomal membranes to generate PtdIns(3)P that regulates membrane trafficking processes via its ability to recruit a ...

Mechanisms of Cellular Membrane Reorganization to Support Hepatitis C Virus Replication

open access: yesViruses, 2016
Like all positive-sense RNA viruses, hepatitis C virus (HCV) induces host membrane alterations for its replication termed the membranous web (MW). Assembling replication factors at a membranous structure might facilitate the processes necessary for ...

Phosphatidylinositol 4‐phosphate dynamics during phagocytosis

open access: yesThe FASEB Journal, 2015
Phosphoinositide levels undergo acute changes during the course of phagocytosis; phosphatidylinositol 4,5‐ bis phosphate [PtdIns(4,5)P 2 ], phosphatidylinositol 3,4,5‐ tris phosphate and phosphatidylinositol 3 ...

Editorial: Membrane lipids in T cell functions [PDF]

open access: yes, 2018
Plasma membrane lipids play essential roles in regulating T cell signaling, differentiation, and effector functions. The major lipid species in the plasma membrane are glycerophospholipids, sphingolipids, and sterol lipids.

An HPLC procedure for separating polyphosphoinositides on hydroxylapatite.

open access: yesJournal of Lipid Research, 1990
We describe a method for separation of several phosphoinositides by high performance liquid chromatography (HPLC) for the purposes of identification, analysis, and possible purification of cell phospholipid extracts.
