A new method for quantitative analysis of M13 bacteriophage by atomic force microscopy

open access: yesSynthetic and Systems Biotechnology, 2022
Quantitative analysis is essential for virus research, especially in determining the virus titer. The classical method plaque assay is time-consuming, complex, and difficult for the phages that cannot form apparent plaque on the solid medium. In order to

Comparison of bovine rotavirus strains by plaque assay [PDF]

open access: yes, 1981
peer reviewedUsing plaque assay on MA 104 cells, four strains of bovine rotavirus were compared: the attenuated American strain NCDV, and three virulent strains isolated in Canada (PQ) or in Belgium (S14 and S 77).

Development of an automated plaque-counting program for the quantification of the Chikungunya virus

open access: yesScientific Reports
Chikungunya virus (CHIKV) induces a massive cytopathic effect (CPE) on various cell types. Therefore, the plaque assay, a CPE-based virus titration method, remains the gold standard for quantifying the infectious units of CHIKV.

Infection Counter: Automated Quantification of in Vitro Virus Replication by Fluorescence Microscopy

open access: yesViruses, 2016
The ability to accurately and reliably quantify viral infection is essential to basic and translational virology research. Here, we describe a simple and robust automated method for using fluorescence microscopy to estimate the proportion of virally ...

Hyperosmotic stress induces PARP1‐mediated HPF1‐dependent mono(ADP‐ribosyl)ation

open access: yesFEBS Letters, EarlyView.
Sorbitol‐induced hyperosmotic stress rapidly induces reversible mono(ADP‐ribosyl)ation (MARylation) on PARP1 without the signs of genotoxic signaling. We show that PARP1 autoMARylation is HPF1 dependent and forms hydroxylamine‐resistant O‐glycosidic linkages.

Salmonella lipopolysaccharide‐containing supported lipid bilayers as platforms to study bacteriophage interactions

open access: yesFEBS Letters, EarlyView.
We present robust protocols for the preparation of supported lipid bilayers (SLBs) incorporating either Salmonella smooth LPS or outer membrane vesicles (OMVs). We use a combination of quartz crystal microbalance with dissipation (QCM‐D) and fluorescence microscopy to both characterize the SLBs of various compositions and to probe their interactions ...

Population dynamics of SNJ1 determined by plaque assay and qPCR.

open access: yes, 2016
The cells of Natrinema sp. J7-2 were infected by halovirus SNJ1 with an MOI of 50 for 1.5 h, then samples were taken from the culture system at indicated times, and the titers of samples were detected by plaque assay and qPCR.

Real-Time qPCR as a Method for Detection of Antibody-Neutralized Phage Particles

open access: yesFrontiers in Microbiology, 2017
The most common method for phage quantitation is the plaque assay, which relies on phage ability to infect bacteria. However, non-infective phage particles may preserve other biological properties; specifically, they may enter interactions with the ...

Microbiome−host proteostasis crosstalk—An emerging perspective on mechanisms and interventions toward healthy longevity

open access: yesFEBS Letters, EarlyView.
Proteostasis and the gut microbiota play a key role in shaping host physiology. Microbiota‐derived metabolites, vitamins, and RNA modulate host proteostasis. Findings from model systems, including C. elegans, indicate microbes can either stabilize or disrupt host proteostasis.
