Development and Validation of a Plaque Assay to Determine the Titer of a Recombinant Live-Attenuated Viral Vaccine for SARS-CoV-2

open access: yesVaccines
The COVID-19 pandemic, caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has resulted in more than seven million deaths worldwide. To reduce viral spread, the Israel Institute for Biological Research (IIBR) developed and produced a new rVSV-SARS-CoV-2-S vaccine candidate (BriLife®) based on a platform of a genetically engineered ...

Viral Plaque Assay v1 [PDF]

open access: yes, 2020
This is part 3.6 of the "Study of MAIT Cell Activation in Viral Infections In Vivo" collection of protocols. Collection Abstract: MAIT cells are abundant, highly evolutionarily conserved innate-like lymphocytes expressing a semi-invariant T cell receptor (TCR), which recognizes microbially derived small intermediate molecules from the riboflavin ...

Variables affecting viral plaque formation in microculture plaque assays using homologous antibody in a liquid overlay [PDF]

open access: yesJournal of Clinical Microbiology, 1977
A liquid antibody microculture plaque assay and the variables that govern its effectiveness are described. The assay is based on the principle that low concentrations of homologous antibody can inhibit secondary plaque formation without inhibiting formation of primary plaques. Thus, clear plaques that followed a linear dose response were produced.

Viral Concentration Determination Through Plaque Assays: Using Traditional and Novel Overlay Systems [PDF]

open access: yesJournal of Visualized Experiments, 2014
Plaque assays remain one of the most accurate methods for the direct quantification of infectious virons and antiviral substances through the counting of discrete plaques (infectious units and cellular dead zones) in cell culture. Here we demonstrate how to perform a basic plaque assay, and how differing overlays and techniques can affect plaque ...

A Useful Quantitative Semimicromethod for Viral Plaque Assay

open access: yesExperimental Biology and Medicine, 1973
This paper presents a general method for rapid, easy, and convenient quantitation of virus titers which we have utilized in applications varying from measurement of virus distribution in infection in vivo to assay of neutralizing antibody or antiviral agents.

New low-viscosity overlay medium for viral plaque assays.

open access: yesVirology journal, 2006
Plaque assays in cell culture monolayers under solid or semisolid overlay media are commonly used for quantification of viruses and antiviral substances. To overcome the pitfalls of known overlays, we tested suspensions of microcrystalline cellulose Avicel RC/CLtrade mark as overlay media in the plaque and plaque-inhibition assay of influenza viruses ...

Standardization of Quantitative Plaque-Based Viral Assays for Orthoflavivirus Cacipacoré

open access: yesViruses
Cacipacoré virus (CPCV) is an understudied orthoflavivirus with significant gaps in research that hinders our understanding of its ecology, host range, and potential public health impact. A notable barrier to filling this gap is the absence of standardized methodologies for viral quantification, such as plaque-forming and focus-forming assays.
