ABSTRACT A 30‐year‐old male sought medical consultation due to sudden anisocoria. Clinical examination confirmed Adie's tonic pupil. Subsequently, the presence of segmental anhidrosis with compensatory hyperhidrosis and areflexia established the diagnosis of Ross syndrome.

Engineering a PLP-dependent Mannichase

Pyridoxal 5'-phosphate (PLP)-dependent enzymes are versatile biocatalysts known for their ability to form diverse C-C, C-N, and C-S bonds. Despite this catalytic diversity, a PLP-dependent enzyme capable of promoting an intermolecular Mannich reaction to access α,β-diamino acids has not been described.

Abstract Objective Medical devices are reviewed by the US Food and Drug Administration (FDA) for safety and effectiveness. Class II devices can receive 510(k) clearance if they are “substantially equivalent” to an existing predicate device. This process can be repeated iteratively, leading to a lineage of predicates for newly cleared devices.

A new branch of mammalian vitamin B6 metabolism: AKR1C‐mediated conversion of pyridoxal to pyridoxine and 4‐pyridoxolactone

Pyridoxal 5′‐phosphate (PLP) homeostasis relies on salvage enzymes, yet key metabolic branches remain undefined. We identify AKR1C isozymes as previously undescribed contributors that convert pyridoxal into pyridoxine or 4‐pyridoxolactone through reductase and dehydrogenase activities.
