SILAC Proteomics of Planarians Identifies Ncoa5 as a Conserved Component of Pluripotent Stem Cells

open access: yesCell Reports, 2013
Planarian regeneration depends on the presence of pluripotent stem cells in the adult. We developed an in vivo stable isotope labeling by amino acids in cell culture (SILAC) protocol in planarians to identify proteins that are enriched in planarian stem ...

Human Primordial Germ Cells Are Specified from Lineage-Primed Progenitors. [PDF]

open access: yes, 2019
In vitro gametogenesis is the process of making germline cells from human pluripotent stem cells. The foundation of this model is the quality of the first progenitors called primordial germ cells (PGCs), which in vivo are specified during the peri ...

Cardiac recovery via extended cell-free delivery of extracellular vesicles secreted by cardiomyocytes derived from induced pluripotent stem cells

open access: yesNature Biomedical Engineering, 2018
The ability of extracellular vesicles (EVs) to regulate a broad range of cellular processes has recently been exploited for the treatment of diseases.

Mechanically and Chemically Defined PEG Hydrogels Improve Reproducibility in Human Cardioid Development

open access: yesAdvanced Healthcare Materials, EarlyView.
Synthetic matrix metalloproteinase (MMP)‐degradable polyethylene glycol (PEG)‐based hydrogels are developed to investigate the influence of mechanical and biochemical cues on cardioid development. Matrix stiffness and cell adhesion motifs significantly regulate cardioid formation, chamber morphogenesis, contractile function, and cardioid transcriptome.

Generation of hiPSC lines with fusion tagged thyroid hormone receptor α isoforms to study isoform-specific actions of TRα1 and TRα2

open access: yesStem Cell Research
The role of the nuclear thyroid hormone receptor TRα1 as a mediator of thyroid hormone action on target gene expression is well understood. However, the function of the TRα2 splicing isoform, which does not bind thyroid hormones, remains unexplored.

Pluripotent Stem Cells: Current Understanding and Future Directions

open access: yesStem Cells International, 2016
Pluripotent stem cells have the ability to undergo self-renewal and to give rise to all cells of the tissues of the body. However, this definition has been recently complicated by the existence of distinct cellular states that display these features ...

Transcriptional profiling reveals fundamental differences in iPS-derived progenitors of endothelial cells (PECs) versus adult circulating EPCs [PDF]

open access: yesarXiv, 2020
There are a number of different stem cell sources that have the potential to be used as therapeutics in vascular degenerative diseases. On the one hand, there are so called endothelial progenitor cells (EPCs), which are typically derived from adult blood. They carry the marker CD34, but the true nature and definition of EPCs is still controversial.

Nephron organoids derived from human pluripotent stem cells model kidney development and injury

open access: yesNature Biotechnology, 2015
Kidney cells and tissues derived from human pluripotent stem cells (hPSCs) may enable organ regeneration, disease modeling and drug screening. We report an efficient, chemically defined protocol for differentiating hPSCs into multipotent nephron ...

Engineering Assembloids to Mimic Graft‐Host Skeletal Muscle Interaction

open access: yesAdvanced Healthcare Materials, EarlyView.
This study develops a graft‐host skeletal muscle assembloid model combining neuromuscular organoids with tissue‐engineered constructs. Pre‐seeding decellularized muscles with myogenic cells enhances cell migration and axon invasion from the organoid. The model exhibits regenerative capacity following acute damage, advancing the understanding of human ...
