PARPing for balance in the homeostasis of poly(ADP-ribosyl)ation

Seminars in Cell & Developmental Biology, 2017
Despite more than 50 years of research, the vast majority of the biology of poly(ADP-ribosyl)ation (PARylation) still remains a gross mystery. Originally described to be a part of the DNA repair machinery, poly(ADP-ribose) (PAR) is synthesized immediately by poly(ADP-ribose) polymerases (PARPs, also known as ARTDs) upon DNA damage and then rapidly ...

The role of poly(ADP-ribosyl)ation in the adaptive response

Mutation Research - Fundamental and Molecular Mechanisms of Mutagenesis, 1996
An involvement of the poly(ADP-ribosyl)ation system in the expression of the adaptive response has been demonstrated with inhibitors of the nuclear enzyme poly(ADP-ribose) polymerase. This enzyme is a key component of a reaction cycle in chromatin, involving dynamic synthesis and degradation of variably sized ADP-ribose polymers in response to DNA ...

A role for poly(ADP-ribosyl)ation in DNA methylation

Biochemistry and Cell Biology, 2003
The aberrant DNA methylation of promoter regions of housekeeping genes leads to gene silencing. Additional epigenetic events, such as histone methylation and acetylation, also play a very important role in the definitive repression of gene expression by DNA methylation.

Physiology and pathophysiology of poly(ADP‐ribosyl)ation *

BioEssays, 2001
AbstractOne of the immediate eukaryotic cellular responses to DNA breakage is the covalent post‐translational modification of nuclear proteins with poly(ADP‐ribose) from NAD+ as precursor, mostly catalysed by poly(ADP‐ribose) polymerase‐1 (PARP‐1). Recently several other polypeptides have been shown to catalyse poly(ADP‐ribose) formation.

Inhibition of Poly(ADP-Ribosyl)ation Allows DNA Hypermethylation

2007
This chapter emphasizes that along the chain of events that induce DNA methylation-dependent chromatin condensation, a post-synthetic modification other than histone acetylation, poly(ADP-ribosyl)ation, participates in the establishment and maintenance of methylation-free regions of chromatin.

Chromosomal protein poly(ADP-ribosyl)ation in pancreatic nucleosomes

Canadian Journal of Biochemistry, 1982
When pancreatic chromatin fragments were prepared and resolved in the presence of 80 mM NaCl, endogenous poly(ADP-ribose) polymerase activity was found to be maximal in nucleosome periodicities of four to five units and did not respond to any further increases in nucleosomal architecture.

[45] Overview of poly(ADP-ribosyl)ation

1984
Publisher Summary This chapter provides an overview of poly(ADP-ribosyl)ation. The formation of poly(ADP-ribose) by chromatin enzymes is actually initiated by ADP-ribosylation on chromatin proteins. The ADP-ribosylation reaction is catalyzed by enzymes which transfer the ADP-ribose residue of NAD + to various acceptor proteins.

Poly(ADP-ribosyl)ation in asthma and other lung diseases

Pharmacological Research, 2005
Inhibition of poly(ADP-ribosyl)ation in oxidative stress-related pathologies has recently emerged as a very effective anti-inflammatory intervention in animal models of arthritis, colitis, diabetes and shock. Recent data from three laboratories also support the role of poly(ADP-ribose) polymerase-1 (PARP-1) activation in asthma.

Genomic Maintenance: The p53 Poly(ADP-ribosyl)ation Connection

Science's STKE, 2007
The integrity of the genome in higher eukaryotes, as well as the modulation of its complex structure and functions, is exquisitely regulated. This genomic regulation occurs as a function of time in a very sophisticated and elaborate biological process called cell cycle progression, resulting in cell division, and is also controlled by a highly ...
