Programmable Antigen‐Specific Immunity via Self‐Adjuvanting Nanovaccines Co‐Delivering Immune Modulators

open access: yesAngewandte Chemie, EarlyView.
The self‐adjuvanting vaccine—comprising a covalent conjugate of antigen and adjuvant—was formulated into cationic lipid nanoparticles smaller than 100 nm to facilitate uptake by immune cells, with supplemental adjuvants incorporated to modulate immune responses.

Poly(I:C) coated PLGA microparticles induce dendritic cell maturation

open access: yesInternational Journal of Pharmaceutics, 2009
Microparticles from poly(D,L-lactic-co-glycolic acid) [PLGA] are of steadily rising interest for the delivery of antigens to immune cells and the induction of a long-lasting immune response for vaccination or immunological tumor therapy. However, if the desired vaccine contains only weak antigens and fails to activate the antigen presenting cells (APC),

Total Leishmania antigens with Poly(I:C) induce Th1 protective response [PDF]

open access: yesParasite Immunology, 2017
SummaryOur proposal was to develop a vaccine based on total Leishmania antigens (TLA) adjuvanted with polyinosinic‐polycytidylic acid [Poly(I:C)] able to induce a Th1 response which can provide protection against Leishmania infection. Mice were vaccinated with two doses of TLA‐Poly(I:C) administered by subcutaneous route at 3‐week interval. Humoral and

HDAC6 Mediates Poly (I:C)-Induced TBK1 and Akt Phosphorylation in Macrophages [PDF]

open access: yesFrontiers in Immunology, 2020
Macrophages are derived from monocytes in the bone marrow and play an important role in anti-viral innate immune responses. Macrophages produce cytokines such as interferons and IL-10 upon viral infection to modulate anti-viral immune responses. Type I interferons (IFNs) promote anti-viral defense.

Automating Vascular Biology: An End‐to‐End Automated Workflow for High‐Throughput Blood Vessel‐on‐a‐Chip Production and Multi‐Site Validation

open access: yesAdvanced Healthcare Materials, EarlyView.
AngioPlate384 is a 384‐well open‐top platform that automates production of more than 100 miniaturized, perfusable blood vessels embedded in hydrogel and supported by stromal cells. Stromal‐endothelial co‐culture strengthens blood vessel barrier function and yields responses useful for translational planning. Scalable and automation‐ready, it suits drug

Pre‐Encoded IFN‐I Sensitivity Exacerbates Memory T Cell Senescence in Solid Tumors

open access: yesAdvanced Science, EarlyView.
Type I interferon (IFN‐I) signaling promotes p21‐dependent cell cycle arrest in senescent tumor‐specific memory T cells, resulting in poor proliferative responses and solid tumor regression during cancer vaccination. Conversely, IFNα/β receptor blockade reinvigorates T cell proliferation to regress solid tumors and is more effective with increasing ...

TBK1 Induces the Formation of Optineurin Filaments That Condensate with Polyubiquitin and LC3 for Cargo Sequestration

open access: yesAdvanced Science, EarlyView.
Phosphorylation of Optineurin by TBK1 induces the formation of filaments that condensate upon binding to linear polyubiquitin. Membrane‐anchored LC3 partitions into these condensates, suggesting that phase separation of filamentous Optineurin with ubiquitylated cargo promotes the sequestration of cargo and its subsequent alignment with LC3‐positive ...

Redefining Dendritic Cell Vaccines: Synergistically Co‐priming DC and B Cells With Nanoparticles Loading Whole Cell Antigens Maximizes the Efficacy of DC Vaccines

open access: yesAdvanced Science, EarlyView.
Dendritic cells (DC) vaccine is an important type of vaccines. However, the limited efficacy of DC vaccines hinders their widely application. This study redefines DC vaccines and provides a new DC‐BC vaccine that have much better therapeutic efficacy.

Poly (I:C) transfection induces mitochondrial-mediated apoptosis in cervical cancer

open access: yesMolecular Medicine Reports, 2016
Polyinosinic acid:polycytidylic acid, known as poly (I:C), is an analogue of double‑stranded RNA, which exhibits direct antitumor effects against several types of cancer. The present study aimed to evaluate the role of poly (I:C) in the apoptosis of cervical cancer cells.
