Lactylation Enhances the Activity of Lactate Dehydrogenase A and Promotes the Chemoresistance to Cisplatin Through Facilitating DNA Nonhomologous End Junction in Lung Adenocarcinoma

Lactylation, linked to glycolysis, is poorly understood in regulation. It is found that LDHA is lactylated at Lys81/Lys318 by AARS1, boosting its activity and cellular lactylation via positive feedback in LUAD. Lactylation regulates NHEJ‐related proteins; their delactylation impairs NHEJ and increases cisplatin sensitivity, offering anticancer insights.

The Crosstalk Between CRL5 and APC/C E3 Ligases Regulates Metastasis and Chemosensitivity of Cancer Cells

This study uncovers a previously unknown crosstalk between two major ubiquitin ligases, CRL5 and APC/C. The authors identify APC11 as a binding partner of CUL5, showing that their interaction regulates neddylation, substrate stability, and mitotic progression.

USP11‐PGAM5 Axis Promotes Neurotoxic Astrocyte Reactivity by Aggravating the mtDNA‐cGAS‐STING Pathway After Intracerebral Hemorrhage

This study reveals that the deubiquitinase USP11 stabilizes PGAM5, triggering neurotoxic astrocyte transformation after intracerebral hemorrhage. PGAM5 promotes mPTP opening and Drp1 dephosphorylation, synergistically amplifying mtDNA leakage into the cytosol and leading to cGAS–STING hyperactivation.
