We have generated temperature-sensitive lethal point mutations in the small nucleolar RNA-associated protein fibrillarin (encoded by the NOP1 gene in yeast) and analyzed their effects on ribosome synthesis. The five alleles tested all prevent synthesis of normal ribosomes, but in dramatically different ways.

Modelling Yeast Pre-rRNA Processing
2007In this paper we present a quantified model concerning the synthesis of pre-rRNAs. The chemical kinetics simulation software Dizzy has been chosen as both the modelling and simulation framework of our study. We discuss the validation of the model against the available experimental data and we show some preliminary results obtained from the study of our

The nucleolus is the ribosome biogenesis center. The nucleolar structure is disrupted upon entry into mitosis and is formed in early G1 phase. To understand the molecular mechanisms of nucleolar assembly and disassembly, we have studied the mechanism of association between factors involved in pre-ribosome RNA (rRNA) processing and rRNA gene chromatin ...

Recognition signals for mouse pre-rRNA processing
Molecular Biology Reports, 1983In order to identify signals for rRNA processing in eukaryotes, mouse pre-rRNA sequence features around four cleavage sites have been analyzed. No consensus sequence can be recognized when the four boundary regions are examined. Unlike mature rRNA termini, distal sequences of precursor-specific domains cannot participate in stable duplex with adjacent ...

Making Ribosomes: Pre-rRNA Transcription and Processing
2014Ribosomes are essential, intricate cellular machines that translate mRNA into protein in all cells. The process of building a ribosome is highly complex. The ribosomal RNA (rRNA) has to be properly synthesized, processed, and assembled with ribosomal proteins in the cell nucleolus.

The role of the 3′ external transcribed spacer in yeast pre-rRNA processing
Journal of Molecular Biology, 1998We have undertaken a deletion analysis of the 3' external transcribed spacer (3' ETS) in the pre-rRNA of Saccharomyces cerevisiae. A stem loop structure immediately 3' to the 25 S rRNA region is necessary and sufficient for processing of the 3' ETS. This is believed to be by cotranscriptional cleavage by Rnt1p, the yeast homologue of RNase III.

Analysis of nucleolar pre-rRNA processing sites in pea (Pisum sativum)
Science in China Series C: Life Sciences, 2003The location of rRNA processing was analyzed by using in situ hybridization with ITS1 probe and immunolabeling of anti-fibrillarin mAb in pea (Pisum sativum) root pole cells. The results showed that rRNA processing sites were in dense fibrillar components (DFCs) and granular components (GCs), but not in fibrillar centers (FCs). Low doses of actinomycin

Processing of eukaryotic pre-rRNA: the role of the transcribed spacers
Biochemistry and Cell Biology, 1995The 17–18S, 5.8S, and 25–28S rRNA species of eukaryotic cells are produced by a series of nucleolytic reactions that liberate the mature rRNAs from the large primary precursor transcript synthesized by RNA polymerase I. Whereas the order of the cleavage reactions has long been established, until recently little information was available on their ...

Birth of the snoRNPs: the evolution of RNase MRP and the eukaryotic pre-rRNA-processing system
Trends in Biochemical Sciences, 1995The ribonucleoprotein particle RNase MRP is required for the processing of yeast pre-ribosomal RNA (pre-rRNA). A structurally related particle, RNase P, is universally required for processing of pre-tRNA, but in bacteria and archaea also cleaves a site in the pre-rRNA.

Post-mitotic dynamics of pre-nucleolar bodies is driven by pre-rRNA processing.
Journal of cell science, 2012Understanding the relationship between the topological dynamics of nuclear subdomains and their molecular function is a central issue in nucleus biology. Pre-nucleolar bodies (PNBs) are transient nuclear subdomains, which form at telophase and contain nucleolar proteins, snoRNPs and pre-ribosomal RNAs (pre-rRNAs).
