ABSTRACT N6,2’‐O‐dimethyladenosine (m6Am) is a prevalent RNA modification located at the first transcribed nucleotide adjacent to the 5′ cap of mRNAs, where it has been implicated in gene regulation. SETD1A Regulates Glycolysis and Senescence of Nucleus Pulposus Cells via H3K4me3–HELZ2/PPARα‐HIF1α Axis to Drive Intervertebral Disc Degeneration

SETD1A is a key epigenetic regulator in NPCs during IDD. In normal NPCs, it sustains H3K4me3–HELZ2/PPARα–HIF1α signaling to maintain glycolytic energy metabolism and proliferation. In degenerated NPCs, reduced SETD1A disrupts this axis, impairing glycolysis and accelerating senescence, highlighting a promising therapeutic target for IDD.

Covalent Reprogramming of Kinase Binders to Modulate Protein Abundance

Electrophilic remodeling of a broad‐spectrum kinase binder reveals how subtle chemical changes reprogram protein fate. An acrylamide analog of a multi‐kinase binder selectively stabilizes Aurora kinase A (AURKA) by suppressing its ubiquitination, while a short‐linker variant converts this stabilizer into a degrader.

Proximity‐Induced Transfer of a Mass Tag Enables Direct Profiling of Active Matrix Metalloproteases

A proximity‐induced, mass‐encoded activity‐based protein profiling (ABPP) approach transfers a MALDI‐detectable α‐cyano‐4‐hydroxycinnamic acid (CHCA) tag directly to active proteases. This tag enables the direct, multiplexed, and quantitative detection of enzyme activity in complex proteomes without enrichment or purification.
